  • During the fruits development of a new cultivar Taiwan Lemon shaddock(Citrus limonum Grandis Osbeck),relationship between contents,distribution,absorption regularity of seven metal elements and metabolism of active oxygen were investigated. The results were as follows.(1)The main peel growth period,flesh growth period and flesh colour-changed period were from April to May,June to September and October to November respectively.(2)The contents of K,Ca,Mg,Zn,Mn and Fe in peel and flesh decreased with the fruit growth,but the total adsorption quality was opposite. K content was highest of seven. K,Zn and Fe primary in flesh,while Ca,Mg and Mn were dominant in peel. The highest contentration of Cu was observed in young fruits and the contents of Cu in peel and flesh were almost the same.(3)Absorption centralization of K,Ca,Mg,Zn and Mn were from July to September,however,

  • Induction of disease resistance is one of the important strategies of biological control on fruit postharvest decay. The mandarin(Citrus reticulata Blancdo‘Shiyueju’)fruit were inoculated with Penicillium digitatum after vacuum infiltration of Benzothiadiazole(BTH). Results showed that the expression of CR-CT1 and CR-BG3 substantially increased 12 h after BTH treatment. The morbidity and lesion area of green mold in mandarins,and the in vivo disease incidences of the fruit were signicantly reduced by different concentrations of BTH. BTH treatment induced the activities of CT,GUN,PAL,PPO,POD,and evoked the increase of H2O2 content. The results suggested that strong expression of CR-CT1 and CR-BG3,enhanced activities of several defense enzymes and H2O2 content are all involved in the induction of disease resistance by BTH treatment in mandarin fruit.

  • In order to evaluate genetic diversity and relationship of yellow flesh peach germplasm,116 yellow flesh peach varieties,as well as other 19 peach germplasms,were used as plant material. A total of 206 alleles were amplified when screened by 28 SSR primers(26 of them distributed on eight linkage groups on Prunus reference map). Among all the alleles amplified,159 showed polymorphic with a percentage of polymorphic bands(PPB)of 76.56%. For yellow flesh peaches,two main groups were formed in dendrogram at a similarity coefficient of 0.82,when clustered by NTSYS using UPGMA method. Bred varieties from America,Europe and Asia were clustered into one group(Ⅰ),and local varieties from China were clustered into another group(Ⅱ),except‘Xi’an Xingrangtao’and‘Tianjin Huangrou’,which were clustered into bred group. When similarity coefficient was 0.87,bred varieties(Ⅰ)were divided into 8 subgroups. Bred varieties from America were distributed in all the 8 subgroups,and bred varieties from Europe were distributed in three subgroups(Ⅰ-2,Ⅰ-3,andⅠ-4),while bred varieties from Asia were only distributed in two subgroups(Ⅰ-3 andⅠ-4). Varieties with the same parents were closely clustered,and genetic relationship of bred varieties matched well with pedigree in groupⅠ. When similarity coefficient was 0.87,Chinese local yellow peach varieties were divided into 7 subgroups. Local yellow peaches from southwest China were mainly distributed in subgroup Ⅱ-1 and Ⅱ-5,and those from northwest China were mainly distributed in subgroup Ⅱ-2,Ⅱ-4 and Ⅱ-7. For variety population genetic relationship,bred variety from America was the nearest population to that of Asia,followed by that of Europe. Chinese local yellow peach was far from bred varieties on population dendrogram.

  • The regulative sequence(1 711 bp)of chalcone synthase gene promoter(CHSP)from Ginkgo biloba L. was cloned by genomic walking. In silico analysis suggested that the sequence contained several typical cis-acting elements,including UV/blue light responsive elements,phytohormone responsive elements,fungal elicitor responsive elements,MYB binding site,TATA-box and CAAT-box. A 1 402 bp promoter sequence upstream 5′ of translation start site of GbCHS was cloned and designated as GbCHSP,respectively. pBI121 + CHSP and pBI121-35S were constructed and transformed into tobaccos by LBA4404. These results showed that pBI121 and pBI121 + CHSP both could drive the transient expression of GUS in tobaccos and pBI121 + CHSP expressed differentially in root,stem and leaf tissues of tobacco. These results will be help to understand the transcriptional regulatory mechanism on GbCHS expression and flavonoids accumulation.

  • To investigate the function of the MaWRKY1 gene,a transcription factor previously cloned from banana fruit,the polyclonal antibody of MaWRKY1 was prepared. The recombinant plasmid pET-MaWRKY1 was constructed with 699 bp segments started from the 168 amino acid to the 400 amino acid including the two conserved motif WRKYGQK and an about 26 kD expressed His-MaWRKY1 fusion protein was identified by SDS-PAGE following the expression of the recombinant His-MaWRKY1 fusion protein in E.coli BL21(DE3)cells with high efficiency. About 0.5 mg • mL-1 fusion protein was obtained after puified by Ni-NTA agarosecolumn,separated by SDS-PAGE,excised from the gel and electroeluted,then the purified fusion protein was used as antigen to prepare polyclonal antiserum in rabbits. After the fifth injection of antigen,the antiserum was obtained and further purified with method of immuno-precipitation. Indirect ELISA test and the western blot analysis showed that the obtained anti-MaWRKY1 polyclonal antibody have good sensitivity and high specificity with a titer of 1︰160 000. Furthermore,western blot assay revealed that a 26 kD peptide was specifically detected in the total proteins of different banana tissues with anti-MaWRKY1 polyclonal antibody,indicating that the antibody could react to the total proteins expressed in banana specifically. In addition,low temperature could induce the expression of MaWRKY1 protein in banana fruit,suggesting that the expression of MaWRKY1 protein in banana fruit was likely to be involved in regulating the chilling-tolerance of fruit.

  • The effects of Ca2+ on the changes of soluble protein,malondialdehyde(MDA)contents,superoxideanion()production rate,superoxide dismutase(SOD),peroxidase(POD)and catalase(CAT)activities in cucumber(Cucumis sativus L.‘Jinchun 2’)seedlings under 50 mmol • L-1 NaCl salt treament was studied. The results showed that the contents of soluble proteins and MDA, production rate,antioxidant enzyme activities were higher in salt stress than control;In salt + Ca2+ treatment,soluble proteins content,SOD,POD and CAT activities increased,while MDA content and  production rate decreased. Under salt stress and salt + Ca2+,the MDA content, production rate raised,while soluble proteins content,SOD,POD and CAT activities declined. The results indicated that Ca2+ could raise the activities of antioxidant enzymes and decrease the membrane lipid peroxidation for cucumber seedlings,which could alleviate the damage of salt stress and improve the salt stress adaptability of cucumber seedlings.

  • A forward and a reverse suppression subtraction hybridization(SSH)cDNA library with 472 and 124 positive clones were constructed respectively with cDNA from eggplant(Solanum melongena)parthenocarpic line D-10 ovary as the tester and cDNA from un-parthenocarpic line 03-2 ovary as the driver. After removing repeat and redundancy sequences,384 expressed sequence tags(ESTs)were obtained. Protein homology search in non-redundant protein database revealed that,under E-value less than or equal to 1e-10,121 ESTs were highly homologous with known proteins,and 136 ESTs could be new genes. The results of gene ontology showed that 107,102,119 ESTs were involved in biological process,molecular function and cell component respectively. Five parthenocarpic related ESTs were obtained,including two MADS-box family,one auxin growth promoter protein,one cytochrome P450 NADPH-reductase and one MAPKK family.

  • A new anther preferential bHLH transcription factor was cloned using a down-regulated expression EST sequence as a querying probe. The gene as BoBHLH1(GenBank accession number:GU390530)was designated. BoBHLH1 is a new member of bHLH transcription factors family related to OguCMS and contains an 309 nt of continuous complete open reading frame(ORF)encoding a poly- peptide of 102 amino acids. BoBHLH1 is preferentially expressed in cabbage anther and have two expression peaks in early and late stages of anther development. The results provide new clues for further understanding of the nuclear-cytoplasmic interaction mechanism of OguCMS.

  • We observed the floral development of anemone and non-anemone type chrysanthemum as well as the histological structures of different types of petals by paraffin section method. The results indicated that the flower bud differentiation of chrysanthemum included two stages such as the inflorescence and floret differentiation. The difference of floral development between anemone and non-anemone type chrysanthemum occurred in the stage of the elongation of floret corolla. The histological structure of disk floret in the anemone type chrysanthemum plays similarities with that of the ray floret not only at their developmental stage but also at the mature stage. They all consist of multiple cell layers including upper and lower epidermal cell layers and internal loose mesophyll cells. Nevertheless,the disk floret of the non-anemone type chrysanthemum just has the petal layout of upper and lower epidermal cell layers. In the development of the disk floret in anemone type chrysanthemum,the exuberant division activity of the internal cells of the petal and the well developed vascular tissues are probably the important reasons for the formation of the anemone.

  • Pollinium morphology of five most common cultivated orchid genera(Orchidaceae)were examined with SEM(scanning electron microscopy). The results showed that thousands up to ten thousands of pollen grainsare aggregated into distinct pollinia. The pollinia presented diversity in shape,from near pear-shaped,near ovate,to oblong oval. The figures of pollen grains are irregular. The tetrad pollen grains were only found in species of Cymbidium ensifolium. Except C. faberi,there are not obvious sticky semiliquid substance covered the pollen grains. The pollinium of the speces studied can be divided into four types based on the sculpturing of exine. There are psilate,near psilate or very obscurely fossete,rough and fossulate or foveolate. There are no aperture on the exinous lay of pollen grains. The pollinium size and sculpturing of exine showed obviously different among the genera and species in this study. Within the genus Cymbidium,the sculpturing of pollinium differed on the species level but not obviously on the cultivated variation level.

  • The lotus(Nelumbo nucifera Gaertn)seed defensin gene was identified from the lotus cDNA library of middle development stage seed embryo. The 561 bp full length cDNA have a 234 bp open reading frame which encoding a 77 amino acid peptide and was designated as NnDefensin. The amino acid sequence of NnDefensin showed relatively high match with other plant defensin genes and phylogeny analysis showed that NnDefensin have close relationship with defensin from Oryza sativa japonica group,defensin precursor from Triticum aestivum and defensin from Plantago major. NnDefensin have a strictly conserved γ- thionin domain of eight cysteine which is characteristic in plant defencin. Specific primers containing restriction enzyme site of SmaⅠ and SacⅠ was used to amplify the sequence of NnDefensin by polymerase chain reaction(PCR). The binary expression vector pBI121 and vector pAhOleo17.8︰GUS which containing a seed specific promoter were digested by the corresponding restricted enzymes respectively,and linked with the NnDefinsin fragment directionally. The resulting construction was obtained and named pBI121-NnDef in which NnDefensin was driven by 35S promoter,and pAhOleo-NnDef in which NnDefensin was driven by seed specific promoter AhOleo17.8 from peanut(Arachis hypogaea L.). The vectors could be transformed into plants mediated by Agrobacterium tumefaciens for the following up research such as gene functional identification and gene transformation for pathogen resistant improvement in transgenic plants and seeds.

  • Young leaves of several rare germlasm resources in litchi and longan were used to observe chromosome numbers and identify their ploidy after pretreated with the mixture of 1︰1 aqueous solution of saturated paradichlorobenzene and 2 mmol • L-1 8-hydroxyquinoline,fixed by Carnoy’s fixative,hydrolyzed with HCl and enzymes,and stained in improved Carbol-fuchsin. The results showed that ‘Lieye’longan was a haploid(2n = x = 15)and grew normally. The parents used to create three mapping groups,including litchi parent ‘Maguili’ and paternal ‘Jiaohesanyuehong’,and longan parent‘Fengliduo’,‘Shixia’and paternal‘Dawuyuan’,‘Zhongqiu 1’were all diploids(2n = 2x = 30). The intergeneric hybrids(A and B)between Litchi and Dimocarpus were also diploids and could grow normally. Moreover,several germplasm resources with distinct trait including litchi‘Ziniangxi’,‘Wuheli’,‘MS-07’,‘SGA’,‘YA1’and‘YN4’as well as a longan‘Shuangji’were all diploids.

  • Plantlets in vitro of three clones,which were previously selected out for their high salt (NaCl)tolerance,were investigated under various levels(0,10,20 and 30 mmol • L-1)of NaHCO3 and NaCl for their growth parameters,shoot length,root length,shoot fresh weight,root fresh weight,shoot dry weight and root dry weight. With increase in the level of NaCl,values of the six growth parameters for the three diploid potatoes were increased,and then deceased. However for NaHCO3,they were kept on to be deceased. The values of growth parameters under NaHCO3 stress were dropped more compared with NaCl. For the same clone,NaHCO3 would do more harm than NaCl under the same concentration.

  • The effects of cold stress on growth index,freezing injury index and chlorophyll fluorescence parameters in seven different genotype cauliflowers with different or same maturity were studied. The changes of chlorophyll fluorescence parameters in different genotype cauliflowers with same maturity under freezing injury stress were also examined. The correlations among growth,cold tolerance and chlorophyll fluorescence parameters were analyzed in cauliflower varieties with different or same maturity. The results showed that there were significant differences among the varieties with different maturity in freezing injury index and minimal fluorescence(Fo),and there were significant differences among the varieties with the same and different maturity in head weight,vegetative mass weight,SPAD value and conversion efficiency of primary light energy(Fv/Fm). Significant differences existed among the varieties with same maturity under freezing injury stress. Correlation analysis of growth index,freezing injury index and chlorophyll fluorescence parameters in different genotype varieties showed that freezing injury index which directly reflects cold tolerance were negatively correlated with SPAD and Fv/Fm. Head weight and vegetative mass weight which indirectly but externally reflects cold tolerance,were positively correlated with Fv/Fm in varieties with both same and different maturity,and positively correlated with SPAD only in varieties with different maturity. In conclusion,conversion efficiency of primary light energy closely related to cold tolerance,Fv/Fm could be used as index of cold tolerance identification in cauliflowers with both same and different maturity.

  • The effects of different concentrations of sucrose and PP333 on in vitro conservation of Chrysanthemum yoshinaganthum were investigated,and the genetic stability of the plants regenerated from the in vitro preserved germplasm was tested too. The results showed that plantlets can be conserved in vitro for > 360 days in the medium MS + KT 2.0 mg • L-1 + NAA 0.1 mg • L-1 + agar 6.5 g • L-1 supplemented with 15–30 g • L-1 sucrose and 6–9 g • L-1 PP333 under temperature(23 ± 2)℃,illumination intensity 2 000–3 000 lx and 12 h • d-1 photoperiod. The survival rate of plantlets was 93.53%–100%. The plantlets grew well after growth recovery. There were no significant differences in the morphology,peroxidase isoenzyme patterns and ISSR-PCR amplification profiles between the plants regenerated from in vitro preserved plants and non-preserved control,suggesting in vitro conservation didn’t affect the genetic stability of the germplasm.

  • Function components contribute to the health care functions and nutritional quality of Chinese jujube(Ziziphus jujuba Mill.),and its processing products. It is necessary to make a systematic study on this subject in order to get further understanding of Chinese jujube’s functional characteristics,offer useful information for developing functional food and improving the health care function of processing products. The major emphasis of this review is the related investigations at home and abroad on Chinese jujube’s functional components(such as flavonoids,three-terpene compounds,polysaccharides and cAMP)and health functions. Some research perspectives of Chinese jujube and its healthy foods were suggested with the purpose of providing useful information for the further development of Chinese jujube industry.

  • ‘Chunmi’is an early maturing peach cultivar bred by artificial pollination. The flower is showy with much pollen. It’s self-fruitful and the yield is high and stable. Its fruit is near round,weights 135–162 g,and fully covered by rose red when ripening. Its flesh is white,melting,with 11.5% soluble solids content and 0.44% total titratable acid content. The stone is cling. The fruit development period is about 70 days. It ripens in early June in Zhengzhou,China.

  • ‘Shennong Xiangfeng’is a mid-early maturing grape cultivar derived from the self progenies of ‘Zizhenxiang’. The berry is obovate,with purple-black color. Its fruit cluster weight is 480 g,and the average single berry weight is 9.7 g. The soluble solids and titratable acid contents are 18.8% and 0.58% respectively. The berry has excellent quality with strong fragrance and sweet taste. It has a high yield,early bearing and a extreme disease resistance. The ripening period of berry is from late August to early September in Shenyang.

  • ‘Linguan’is a new processing chestnut cultivar selected from Chinese chestnut population growing in Taihang Moutain in Xingtai. It had some good characteristics,such as prematurity,high yield and strong resistance. Size of nut is big. The average fruit weight is 11.91 g. Kernel clour is yellow. The flesh is fine and sticky. The nut is perfect for processing peeled roasted chestnuts and kernel can. It bears fruits in the second year after grafting. The yield of per square meter crown projection area is 1.77 kg. The suitable area is thickness layer and fertility sandy loam or sandy soil weathered by gneiss.

  • ‘Shiguang’is a new fresh Chinese jujube(Ziziphus jujuba Mill.)cultivar selected from clones of‘Linyi Lizao’. The fruit is oblate. The average fruit weight is 34.3 g,the maximum is 108 g. The flesh is juicy,sour-sweet,with 14.48 kg • cm-2 fruit hardness,26.3% soluble solids content.‘Shiguang’ has excellent characteristics such as fine flesh,high resestance to fruit cracking,high yield,early bearing. The fruit developing period is 105–115 days.

  • ‘Sujiao 15’is a new mid-early ripening hot pepper hybrid. Its fruit is long cattle-horn shaped,green color in immature and red color in mature,18–20 cm in length and 96.9 g in weight. The marketable yield is up to 44 000 kg • hm-2. It shows good resistance to disease,excellent storage capacity,and strong tolerance to low temperature. It is suitable for the protected cultivation.

  • ‘Bilü Cutai Caixin’which cultivated by systematic breeding is an early maturity flowering Chinese cabbage cultivar with tolerance to flood and hot. The leaves are oval,oil green,16.2 cm in length and 8.8 cm in width. Main sprouts are 18.4 cm in high,1.5 cm in thick. The single sprout weight is 25.0 g. Sprouts color is oil green. It taste crisp and sweet. The souble solids content is 4.3%,crude protein content is 2.8%,reducing sugar content is 0.95% and vitamin C content is 1 350 mg • kg-1.‘Bilü Cutai’showed strong adaptability,mudium resistance to hot and downy mildew,anthracnose and soft rot. It is suitable for producing in autumn in Guangdong Province.

  • ‘Huayu’is a new watermelon hybrid cultivar. It is early maturing. Its growth and disease resistance are mild strong. Fruit-setting is excellent. The fruit is long-round or oval in shape. Its smooth skin is green with dark green longitudinal stripes. It has thin peel,red and crispy flesh,small and brown seeds. The weight of single melon is 2.3–3.0 kg. The center sugar content of the fruit is about 11%. The fruit maturing stage is 28–30 days.

  • A new Lilium oriental L. cultivar‘Guiyang Red’which is selected from the cross‘Caiyun 3’בRuyi’. The new cultivar is stable. The flower color is bright red with white edge. The stigma color is milk white. The stem color is green with purple spots. The cultivar strongly resist to Fusarium,which has strong adaptability and easy propagation. The new cultivar is an excellent cut-flower.

  • ‘CAF4’is a new Cajanus cajan cultivar,bred from offspring of hybrid pigeonpea cultivar‘ICPH3432’which was introduced from India. Single plant selection and group selection were applied into the breeding course. Its growth period ranges 162–166 days and average number of pod per plant is(644.4 ± 202.4);Each pod contains(3.6 ± 0.4)beans;100-seed weight is(9.62 ± 0.89)g;Total seed weight per plant is up to(113.76 ± 41.64)g.
