  • The effects of summer shoot growth on sugar metabolism and abscission of fruitlet in ‘Shatangju’mandarin(Citrus reticulata Blanco)were investigated. The results showed that, summer shoot obviously increased fruitlet abscission and suppressed fruitlet growth. During 18 days of summer shoot growth, the relative abscission rate(RAR)tended to increase and subsequently decrease, with the peak of 7.63% · d-1 at the 9th day. At the 18th day,the accumulative abscission rate(AAR)in shoot-retained treatment was 94.05%,which was 1.58 times higher than that in shoot-removed treatment. Moreover, summer shoot growth decreased the contents of sucrose and reducing sugar in fruitlet at 6th day,but no significant difference was observed for those between shoot-retained and shoot-removed treatments at 18thday. Furthermore,it was found that summer shoot increased the activities of acid invertase(AI)and neutral invertase(NI),and inhibited those of the amylase and sucrose phosphate synthase(SPS)in fruitlet. However, sucrose synthase(SS)activity was not influenced by summer shoot. Overall, these findings suggest that the accelerated decomposition of sucrose by the increased activities of sucrose degradation enzymes during summer shoot growth contributed to fruitlet abscission in‘Shatangju’mandarin.

  • To make clear the physiological responses of apple trees grafted on three rootstocks under high level of manganese stress. The physiological responses and manganese accumulation of three rootstocks on 400 mg · kg-1 manganese stress were studied. The results showed that toxic symptom happened in new leaves of all apple trees grafted on three rootstocks, which was the most serious in Malus baccata(Linn.)Borkh.,followed by Liaozhen 2 inter stock, the most lightly in Malus prunifolia(Willd.)Borkh. Under 400 mg · kg-1 Mn stress, the growth of apple trees on Malus baccata(Linn.)Borkh. and Liaozhen 2 inter stock were restrained significantly, but the growth of apple trees and Mn accumulation in underground parts on Malus prunifolia(Willd.)Borkh. were promoted, the contents of ROS and MDA were declined sharply, the SOD activity and AsA content increased slightly in apple leaves on Malus baccata(Linn.)Borkh. and Liaozhen 2 inter stock. There were no significant difference of ROS content and a little rise of MDA content because of the much larger increases of antioxidant enzymes of SOD,POD,CAT and antioxidants such as AsA under the high level of Mn stress. The rootstocks of Malus prunifolia(Willd.)Borkh. had higher endurance to manganese toxcity, and Malus baccata(Linn.)Borkh. and Liaozhen 2 inter stock were relatively lower.

  • Effects of different bagging treatments on the soluble sugar contents and sucrose metabolism of ‘Chili’(Pyrus bretschneideri Rehd.)fruit during growth and development were studied. The results showed that the contents of fructose, glucose, sucrose and total soluble sugar in the fruits increased with the fruit growth and development. Bagging, especially with yellow waxed paper bags, reduced the soluble sugar increases. Invertase(Ivr)activity decreased with fruit development, and bagging affected the acid invertase(AI)activity. AI activities in fruits of bagging treatments were lower than that in control in 75,90 and 150 d after flowering. The neutral invertase(NI)activity showed no significant difference between bagging treatments and control. Sucrose phosphate synthase(SPS)activity increased with the fruit development and affected markedly by bagging. Among 4 tested bags, the yellow waxed paper bags affected SPS activity of fruits more than the white waxed paper and non-woven fabric bags did,and there were not significant different between fruits in plastic film bags and non-bagging control in most of the tested points. Sucrose synthase(SUS)in synthesis activity(SSs)of fruits increased gradually and sucrose synthase in cleavage activity(SSc)decreased as fruit growth. Bagging did not showed significant effect on the SSs activity in most tested points, but depressed the SSc activity in fruit development late stage. Real-time fluorescent quantitative PCR analysis of sucrose metabolism related enzyme genes AIV1AIV2SPS1 and SUS1 expression showed that bagging reduced the relative expression of AIV2,and non-woven bags had more effects than plastic film bags. Bagging delayed the maximum accumulation time of SPS1,and the SPS1 relative expression were significantly lower in fruits with plastic film bags than in non-bagging control in the fruit growth early stage. Bagging altered the time course of SUS1 expression, the relative expression amount of SUS1 in fruits with plastic film bags did not reveal obvious peak, and in fruit with yellow waxed paper bags appeared a maximum accumulation 15 d earlier than control and became lower after.

  • Following interspecific hybridization between Actinidia chinensis var. diliciosa‘Xuxiang’and A. longicarpa, pollen tube behaviour among different crossing combinations was determined using fluorescence microscope, difference in fertilization process and embryo development were observed by conventional paraffin sectioning, and fruit-setting rate and hybrid seed quantity were investigated as well. The results showed that the pollen grains of A. longicarpa which is used by interspecific hybridization germinated and grew on the papillate stigma of‘Xuxiang’,but the pollen tubes grew slower than those of the control,wave-like pollen tubes, growth stop and random deposition of callose along pollen tube wall were observed. Fertilization process delayed, embryo grew abnormally and the fruit-setting rate of interspecific hybridization was lower. The seed number in fruit was less than that of introspecific hybridization or open pollination, but there was no obvious difference in the quality of regular seed among crossing combinations.

  • Solanum lycopersicoides showed high resistance to Cucumber mosaic virus(CMV). An introgression line(IL) population derived from a nightshade related species S. lycopersicoides LA2951 was used to identify the involved loci for resistance to CMV by rub-inoculation in four independent experiments. In total,8 QTLs for resistance to CMV located on chromosome 2,7,8,9,10,11 and 12 were found respectively. Percentage of disease index(PDI)was significantly decreased in these ILs. All QTLs were new loci when compared those identified previously from S. habrochaites LA1777 and S. chilense LA0458. The genetic analysis of these QTLs showed incompletely dominant. The interaction of these QTLs presented complete additive effect. The results provide a prospect for breeding of CMV resistance in tomato.

  • Melting temperature(Tm)was different according to the different lengths or nucleotide sequences of DNA fragments. In this report,we established a novel single nucleotide polymorphism(SNP)genotyping system refered as CAPS-MC approach. The CAPS-MC approach combined cleaved amplified polymorphic sequence(CAPS)with melting curve(MC)analysis. By aligning whole genome re-sequencing data from two Brassica campestris accessions,‘L58’and‘ZCT095’,to the reference genome sequence of‘Chiifu-401-42’,a total of 719 HindⅢ endonuclease recognition nucleotide sequence SNP polymorphisms were converted to CAPS-MC markers. We established and optimized CAPS-MC system by randomly selecting 10 pairs of these markers. Our results clearly demonstrated that CAPS-MC approach was sensitive, accurate, rapid and inexpensive. Because the melting curve of 96 samples can be measured in 22 mins once,it is well suited for high-throughput genotyping.

  • For an intergeneric cross between Sinocalycanthus chinensis and Calycanthus floridus var. oblongifolius,a pollen-pistil interaction using fluorescence microscopy and hybrid embryo development through a paraffin section were conducted to reveal intergeneric cross-compatibility. Results showed that, with or without reciprocal crosses, pollen grains of male parents could adhere and further germinate on stigmas of female parents and that pollen tubes could reach the embryo sacs. The process of pollen germination and pollen tube growth was more consistent when S. chinensis was used as the female parent with most of the pollen tubes entering the embryo sacs approximately 36 h after pollination. However, with or without reciprocal crosses, the hybrid embryo development through the paraffin section caused post-fertilization barriers that led to a very low seed set. Although double fertilization was observed, most hybrid embryos aborted in the early developmental stage. The barrier mechanism between reciprocal crosses, though, differed, with the reproductive barrier for S. chinensis × C. floridus var. oblongifolius being due to hybrid embryo abortion or degeneration in the early developmental stage, and the reproductive barrier for C. floridus var. oblongifolius × S. chinensis being due to a high percentage of abnormal megagametophytes together with hybrid embryo abortion in the early developmental stage. Thus the cross between S. chinensis and C. floridus var. oblongifoliushad no barriers from pollen adherence and germination, but post-fertilization barriers meant poor seed set.

  • The composition and content of anthocyanins in the petals of Camellia reticulata Lindl. were studied shading at the different levels. The results showed that 13 major anthocyanins in the petals were identified by high pressure liquid chromatography(HPLC)analysis. Among which cyanidin 3-O-(2-O-b-xylosyl)-b-glucose (Cy3GX)and cyanidin 3-O-[2-O-b-xylosyl-6-O-(E)-p-coumaroyl]-b-glucose (Cy3GEpCX)were the main anthocyanin in the petals of C. reticulata, and theirs contents reach up to 60.4%–64.2% of total anthocyanins in petals. The contents of the 13 anthocyanins were observed increase at first then reduce with the shade rate increased,and/or the shading time extended, but theirs percentage did not change among total anthocyanins. The contents of Cy3GX,Cy3GEpCX and total anthocyanins were found to increase significantly when the plant was covered by 70% shade net;And 60% shade net covering have similar effects. Comparing with non shading plants, the highest content of anthocyanins was produced at the 21st day covering in the 60% and 70% shade nets. However, the contents of the three anthocyanins were reduced with the shading time was extended in 90% shade net, even lower than the non-shading. It is important for the plant of Yunnan camellia flowersbeing appropriate shaded with shade net in order to increase quantity of anthocyanins, and to improve the flower color.

  • Orthogonal design was applied to optimize SCoT-PCR amplification system of Chrysanthemum in five factors such as Mg2+,dNTPs,primer,Taq DNA polymerase and template DNA. An optimal reaction system of species from Chrysanthemum was completed;25 μL PCR reaction mixtures contained Mg2+ 1.2 mmol · L-1,dNTPs 0.15 mmol · L-1,primers 0.8 μmol · L-1Taq polymerase 1 U,template DNA 50 ng. The most suitable annealing temperature of primers was 49.4 ℃. The improved method for separating PCR products was 8% polyacrylamide gel electrophoresis followed by silver staining. Three materials from Chrysanthemum with different ploidy levels(2x,4x,6x)were used to test the optimized reaction system. Ideal results with clear polymorphic bands showed that the stable and reliable reaction system was obtained. SCoT markers were applied to study the genetic diversity and relationship of 18 species of Chrysanthemum and its related genera. Using 12 selected SCoT primers 209 bands were generate,of which 189(90.43%)were polymorphic. The Jaccard genetic similarity coefficients among the species were calculated with NTSYS-pc2.10e software and the values were between 0.4516 and 0.7035 with an average of 0.58. These coef?cients were utilized to construct a dendrogram using the unweight pair-group method with arithmetic averages(UPGMA). The results showed that 18 species of Chrysanthemum and its related genera could be grouped into two distinct clusters based on similarity of 0.530. The others except Crossostephium chinense and Artemisia sericea were in one cluster, which was further divided into six groups when the similarity coefficient value was 0.615. The results demonstrate that the SCoT marker system is useful for studying genetic diversity and relationships among species of Chrysanthemum and its related genera.

  • During the past 20 years, the pathway of chlorophyll breakdown has been deep- investigated, especially the structure elucidation of nonfluorescent chlorophyll catabolites which is crucial for the cognizance of chlorophyll breakdown pathway. The effects of some catabolic enzymes involved in the chlorophyll degradation have been further demonstrated and part of the pathway is revised and completed. Moreover, the physiologic roles of chlorophyll catabolites enrich the significance of chlorophyll breakdown which is not only a prerequisite to detoxify the phototoxic pigments catabolites, but also is relevant to the viability of senescent leaves and ripening frui.

  • ASR(the abscisic acid, stress, ripening protein)proteins are a suite of important family proteins found in organisms, which are generally involved in osmoregulation. They will be induced in plants under stress conditions, such as drought, low temperature, salt stress and ABA. The overexpression of ASR proteins can reduce plant injure by stress. ASR proteins can stabilize the cell membrane structure, and as molecular chaperones, can combine ions and prevent oxidation. In current paper, the classification and structure of ASR proteins, encoding genes, the expression and regulation of ASR proteins, and the relationship among ASR gene expression,ASR protein accumulation and plant stress tolerance has been summarized.

  • The presence ofRNA molecules in plant phloem sieve elements which can do long-distance transport is an important discovery in plant kingdom. And it has been discovered that there are three kinds of RNA molecules in phloem sap including exogenous viroids or virus RNA,plant endogenous mRNA and small non-protein-coding RNA(snRNA). Long-distance transport of RNAs in phloem sap can function as signal molecules in mediating plant growth and development. In this paper, the methods of how to collect phloem sap,the RNA molecules in plant phloem sap and the signaling pathways as well as the mechanisms by which how they are transported are discussed and summarized.
