Induction and Determination of Tetraploid Plants of Phaius tankervilleae

Abstract: Tetraploids of Phaius tankervilleae were induced through the treatment of protocorms with colchicine in vitro. The results showed that ploidy levels of protocorms changed with colchicine soaking and the highest induction rate of 22.5% was obtained by immersing protocorms into 0.02% colchicine solution for 6 days. Cytological determination of regenerated plants indicated that three tetraploid plantlets were obtained among mutated plants survived,and the morphology,the number and size of stomata of the mutants were different from those of the diploid plants,with obviously bigger nucleus.
