Cloning and Expression Analysis on AP1 Homologous Gene from Cymbidium ensifolium and Interaction Analysis Between AP1 and MADS-box Transcription Factors

Abstract: To investigate the function of AP1 genes on flower development in Cymbidium ensifoliumAP1 was cloned from‘Sijidaqing’flower buds and the full length of AP1 cDNA was amplified by RACE method,named CeAP1. CeAP1 is 866 bp in length with a 726 nucleotides ORF that putatively encodes a protein with 241 amino acids. Homology analysis showed that the CeAP1 was highly homologous to the AP1MADS-box genes of orchids. Quantitative real-time PCR analysis demonstrated that CeAP1 was expressed in the initial period of floral development and vegetative organs. The yeast two-hybrid analysis indicated that the CeAP1was not only able to form homodimer but also formed heterodimers with the other MADS-box transcription factors except for CeAG1.
