Subcellular Localization and Expression Analysis of CsICE Transcription Factor Related to Cold Stress in Tea Plant

Abstract: The 783 bp length ORF of transcription factor CsICE,related to the cold stress in tea plant [Camellia sinensis(L.)O. Kuntze],was amplified by PCR,and cultivar‘Yingshuang’was used as the material. The expression levels of CsICE were analyzed by real time quantitative PCR. The result showed that CsICE expressed in all the tissues,including roots,stems,leaves,flowers and fruits. Furthermore,the expression level in leaves was the highest,almost four times higher than that in roots,stems and fruits. The expression level of CsICE was 40 times under ABA stress(500 μmol ? L-1)higher than that in untreated sample,while under cold treatment at 4 ℃,it was 20 times higher than the control. Under 10% polyethyleneglycol treatment,the CsICE expression level was only 5 times higher than that in untreated sample. The localization assays result indicated that CsICE protein localized in the cell nucleus.
