  • Malus toringoides Hughes. , being complex diversities in morphological characters, germplasms, distributing habitats and resistance to adverse conditions, is an important apple germplasm. There are a number of
different types derived from Malus toringoides. The concept ion of population, pictorial scatter diagrams and hybrid index is adopted to invest igate the variation and differentiation among the plants in M. toringoides , M. transitoria and M. kansuensis . M. toringoides is a species of facultative apomixis. It can produce variations through intro gressive hybridization with M. transitoria and ( or) M. kansuensis , then maintain the new genetic variat ion through
facultative apospory. After millions of years of multiplying, it has become a complicated hybrid population hybrid complex of three species of Malus. Through invest igat ing the rate of gene recombinat ion, the mutation and gene bank of descendants, the genetic mechanism of highly resistance to adverse condit ions could be revealed. In order to maintain and increase diversities of this species, wed better culture M. kansuensis and M. transitoria in populations of M. toringoides .

  • In this study , an AFLP marker of Co gene of apple was converted into a SCAR marker successfully. After sequencing of the AFLP marker fragment , two pairs of primers CoA1/ CoA2 and CoA1/ CoA3 were designed according to the nucleotide sequence. PCR results from apple genomic DNA showed that CoA1/ CoA2 could amplify two bands with the size of 216bp and 148bp respectively , and the one of 216bp was the character band of columnar phenotype ; CoA1/ CoA3 could amplified two bands too , and the size was 273bp and 205bp respectively , the former one was the character band of columnar phenotype. Both of the two pairs of primers have high correct rate in judgment of columnar growth habit ( > 90 %) , so either of them can be served as special primers of this SCAR marker for rapid selection.

  • The paper reported the detection proficiency of the seedlessness in 21 seedless grape cultivars and 9 seeded ones using a specific primer which was a probe available for detecting seedless genes in grapes’genome. The
results showed that the specific probe could be accurately used in the cultivars cultured in China. The preselection of seedlessness was also reached in cross combination‘seeded cultivars (Red Globe ) ×seedless one (Flame seedless) ’progenies. It furthermore demonstrated that the seedlessness in grapes was controlled by multigenes and the specific genetic marker was linked to one of the seedless genes in grapes.

  • The adjustment of leaf/ fruit ratio, girdling and the application of CPPU were taken to investigate the influence of sink or source change on fruit characteristics in Qinmei ( Actinidia deliciosa) . The results showed that the fruit size fluctuated more remarkably than fruit quality and other characteristics after different treatments. The leaf/ fruit ratio 2- 41 in fruiting lateral could meet the normal demand of fruit growth and development. The
competitive ability of fruit not source strength is a major factor determining the final fruit size. The relationship be btween cell number and fruit size as well as that between hardness and Ca content was discussed.

  • The banana (Musa acuminata AAA group) seedlings were sprayed with single or mixed H2O2 and CaCl2 and exposed to 7 ℃low temperature for 24 h. The increase of POD activities and soluble sugar contents and alleviation of cell membranes leakage and chlorophylls decomposition in banana seedling leaves were observed during cold stress and recovery periods. The mixed H2O2 and CaCl2 had a synergism on these aspects. The results also showed that H2O2 had better effects in increasing peroxidase activities and Chl . b contents than those of CaCl2 , but CaCl2 had advantage effects on soluble sugar and Chl . a contents. POD activies had a significant opposite realation with electric leakage and it may suggest that increasing POD activities could help to decrease the leakage of cell membranes during low temperature period.

  • In the experiment, the morphological and physiological changes of tomato at different growth stages under low light were studied. The results showed that the tomato seedlings had a tendency of overgrowing under low
light. Low light at seedling stage led to the increase of SLW, LAR, Chlorophyll Content, Chlorophyll b/ a value and percentage of fruit setting and the decrease of soluble sugar and protein content. While the low light at
blooming and setting stage led to the increase of POD activity and soluble sugar content and the decrease of the percentage of fruit settings, comparing with that at seedling stage.

  • UV irradiated protoplasts ( donor protoplasts) isolated from petaloid cytoplasmic sterile line 7-0-8 were fused with 15 mM iodoacetamide treated protoplasts ( recipient protoplasts) isolated from fertile material 66-3 by using electronic fusion method, 33 plants were regenerated from fusion cells. All regenerated plants were diploid with chromosome number of 18, and they possessed the identical vegetative morphology to the recipient parent 66􀀁3.
In PCR amplification with mtDNA specific primers ( STS4 primer combination) , all regenerated plants obtained one same specific band which was identical to donor parent and different from recipient parent. The results indicated that
all regenerated plants were cybrids. In this experiment, 4 plants identified for their flower morphology were petaloid male sterile, which proved that intraspecific cybrids in carrot were obtained by asymmetric protoplast fusion and car rot petaloid cytoplasmic male sterility was successfully transferred from donor parent line 7-0-8 to fertile line 66-3.

  • DL-β-aminon-butyric acid ( BABA) that was applied as a foliar spray to pepper plants induced resistance to a challenge infection with Phytophthora capsici . A relatively high concentration of BABA at 1 000 g/mL, which had no antifungal activity in vitro against P . capsici , was required to induce resistance against Phytophthora blight with a foliar and stem spray, thus leading to complete control of the disease. About 3 days interval
between BABA􀀁treatment and challenge inoculat ion was sufficient to induce resistance in pepper plants, and the in duced resistance could be retained more than 20 days after BABA􀀁treatment. High inoculum levels of P . capsici
    caused
Phytophthora development slowly in pepper foliar treated with BABA, especially at early plant growth stage, which suggests that the induced resistance in pepper plants may be more quantitative rather than qualitative.

  • An efficient plant regeneration system was established through optimization of hormone combinat ion, genotype, seedling age and root induction medium in root tip culture of Chinese chive. Data showed that the best medium for callus and adventitious shoot differentiation wasMS medium supplemented with 1 mg/ L NAA and 2 mg/L BA. Shoot induction rate of 91-1, 91-8, Baoding Honggen, Shouguang Malinjiu and Lanzhou Xiaojiu were 78. 7%, 83. 7 % , 81. 9 %, 76. 7 % and 73. 3 %, while the average shoot number per explant were 40. 1 , 46. 7 , 36. 3 , 35. 4 and 44. 5 respect ively. Root t ips taken from plantlets 7 to 10 day old seedlings showed the highest regeneration ability. MS medium without growth regulators was optimum for root formation of regenerated shoots with 100% root induction rate and 14. 5 roots per shoot. Kanamycin inhibited callus and shoot induction definitely at 20 mg/ L. Carbenicillin and Cefotaxime had inhibit ing effects on callus induction and shoot regeneration at Carbenicillin 500 mg/ L and Ceftaxime 300 mg/ L respectively. The effect of Timent in was not obviously, with the inhibition effect only lying in the reduction of shoot number induced from callus.

  • The ultrastructure and photosynthetic characters of chloroplast were investigated at setting stage in leaves of glabrous and normal cucumbers. Compared with that of normal cucumber , the structure of photosynthetic
apparatus in glabrous cucumber was simple with less numbers of chloroplast and grana as well as the grana lamellae. The contents of chlorophyll a and b and carotenoid in glabrous cucumbers only reached half of the content in normal one. Therefore , the capacity of capturing light energy was low. The photosynthetic rate of glabrous was obviously lower than that of normal , no more than half of normal cucumber during most times in a day. Diurnal change of photosynthetic rate of glabrous was unimodal distribution with the maximum occurring at 11 :00 , whereas diurnal patterns of normal one displayed bimodal distribution which two peaks occurred at 10 :00 and 12 :00 . The photosynthetic rate of glabrous cucumber increased as CO2 concentration increased , the maximum reached 1300μL·L - 1 of CO2 , but its level was significantly lower than that of normal cucumber under the same CO2 concentration.

  • Abstract ( 1523 ) HTML ( 1182 ) PDF (318KB) ( 1182 )
  • No PAO activity was detected in ungerminated cowpea (Vigna unguiculata L. ) seeds (form 0 to 24 h after imbibition) , but it could be detected in the plumule , cotyledons and embryonic axis of the germinatingseeds whose radicles were just visible (36 h after imbibition) . The PAO activities in these three parts increased at different rates during germination , while no PAO could be detected in radicle at all times. Soaking with Act2D only inhibited the seed germination by 27. 6 % , but CHM could inhibit the germination by 90. 6 %. Moreover , the inhibitions of the PAO activity in the plumule and cotyledons by Act2D were just 29. 1 % and 33. 3 % , and those by CHM were up to 87. 2 % and 85. 4 % respectively. Light had a remarkable inhibitory effect on the PAO activity in plumule and cotyledons. Soaking with putrecine (0. 1 and 0. 5 mmol/ L) or CuCl2 (10μmol/ L) for 6 h could enhance the activity of PAO.
