Abstract: The callus of Cleopatra ( Citrus reticulata) , a citrus rootstock cultivar , was transformed by the strain of Agrobacterium tumef aciens EHA105 carrying gfp gene. The callus of Cleopatra ( Citrus reticulata) was co cultivation with A. tumef aciens and selected with hygromycin , as led to the regeneration of resistant embryogenic callus and later to plantlets. The plants were verified GFP2transgenic plants by GUS analysis , GFP2fluorescent detection , PCR amplification and Southern hybridization.
