Incidence and Molecular Characteristics of Citrus leaf blotch virus and Citrus tatter leaf virus Infecting Citrus in China

Abstract:

Both Citrus leaf blotch virus(CLBV)and Citrus tatter leaf virus(CTLV)can infect citrus species. To understand the infection status and molecular characteristics of the two viruses in citrus plants in China,citrus samples collected from seven provinces were detected by reverse transcription-polymerase chain reaction(RT-PCR). Results showed that the incidences of CLBV and CTLV were 9.6% and 11.0%,respectively. The partial coat protein(cp)gene of 14 CLBV isolates shared 82.1%–100% sequence identities at nucleotide(nt)level and 88.1%–100% sequence identities at amino acid(aa)level,respectively. The partial cp gene of 15 CTLV isolates shared 89.5%–100% nt and 92.5%–100% aa sequence identities,respectively. Phylogenetic analyzes of partial cp gene sequences of both CLBV and CTLV showed that CLBV isolates identified clustered into two groups. The 15 CTLV isolates separated in different branches,and there is no clear relationships between their phylogenetic positions and original hosts.
