Construction of Molecular Linkage Map in Grape Using RAPD Markers and F1 Population of Interspecific Cross

Abstract: A set of 280 randomoligonucleotide perimers was screened for random amplified polymorphic DNAs (RAPD) fragments within a sample of 60 F1 hybrids to generate RAPD markers and construct the molecular linkage map fromtwo parents 8324296 (V. quinquangularis) and Muscat Rose (V. vinifera) . 89 repeatable RAPD markers as well as four morphological markers (flower type , grape downy mildew2resistance , berry color and juice color) were obtained. The map consisted of 11 linkage groups , it covered a total genetic distance of over 1 033 cM (centimorgan) , the average distance between markers was 17.8 cM. It supplied a linkage framework for a more saturated linkage map of grape.
