The Expression of RcHSP17.8 Gene Improving Tolerance to Abiotic Stress inRecombinant E.coli Cells

Abstract: RcHSP17.8 is a high temperature (HT) induced expressing gene, by RT-PCR and Southern blot indicate that there are several copies of RcHSP17.8 in genome DNA of Rosa hybrida L. RcHSP17.8 is more strongly expressed in varieties resistant to HT, such as‘Schloss Mannieim’, ‘Radio’, ‘Las Vegas’,
‘Caprice de Meiland’, ‘Girl’, meanwhile weakly or not expressed in varieties sensitive to HT, such as‘Kordes’Perfecta’, ‘Hiogi’, ‘Pfalzer Gold’. The results indicate that this gene is possibly relative to the
tolerance in Rosa hybrida L. to HT. In order to study its possible function, RcHSP17.8 gene has been transferred into Escherichia coliBL21. Recombinant RcHSP17.8 was overexpressed in the recombinant E.coli cells which showed imp roved viability under different abiotic stress conditions, such as, high salt concentration,heavymetal and oxidative stress, as compared with control cultures. Those resultswould provide molecular identification principle to selecting varieties of Rosa hybrida L. with strong tolerance to HT.
