  • Effects of Bagging on Accumulation of Phenolic Acids and Flavonoids in Peach Pericarp During Fruit Maturity

  • Trail cultivar was‘Wanmi’peach ( Prunus persica L1). Fruits were bagged on 75 days and bag removed on 144 days after full bloom, the nonbagged fruits were as the control. The phenolic acids and flavonoids in pericarp were qualitatively and quantificationally analized by HPLC - MS. Three kinds of phenolic acids, five kinds of flavan-3-ols, eight kinds of flavonols, and two kinds of anthocyanins were isolated,detected and quantified in pericarp of ripe peach. With the ripe of fruits, the contents of phenolic acids and flavan-3-ols in pericarp were gradually declined; The content of flavonolswas high at early ripe stage, it was increased sharply at the early fruit coloring stage, and then sharp ly declined at the late stage of fruit maturation1. Anthocyanins in nonbagged fruits were found to be accumulated on 144 days after full bloom, which were
    accumulated in bagged fruits after removing the bags. The contents of phenolic acids and flavan-3-ols had no obvious difference between bagged and nonbagged fruits when fruits were ripe. The contents of anthocyanins
    and flavonols in the bagged fruits were 1.74 times higher than that in the nonbagged fruits1.The results indicated that phenolic acids and flavan-3-ols were sensitive to light, bagging could restrain the synthesis of them in
    fruits except the ripe fruits, but bagging could enhance the accumulation of anthocyanins and flavonols significantly in the ripe fruits.Three major biosynthates were found in the pathway of accumulating anthocyanins,
    each metabolism branch was existed at the stage of young fruit, flavonols and anthocyanins were accumulated synchronously at the early fruit coloring stage and anthocyanins was accumulated at the fruit ripe stage.

  • By comparing with two protein extraction methods, ensured the optimal method for longan seed proteins extraction. Proteomic approaches were used to analyze the changes of differentially expressed proteins during four different stages of longan seed abortion. Twenty-one differentially expressed proteins were detected on 2-DE gels, 9 were up-regulated and 12 were down-regulated. Among these proteins, 12 proteins were identified by MALDI/TOF /TOF-MS/MS and protein database searching. They were cytosolic malate dehydrogenase, Dihydrolipoamide adetyltransferase, 2, 3-bisphosphoglycerate-independent phosphoglycerate mutase, RuB isCO large subunit2binding p rotein subunit beta chlorop last p recursor, RuBisCO large subunit-binding protein subunit beta chloroplast precursor, 17 kD heat shock p rotein, ascorbate peroxidase, cytosolic ascorbate peroxidase, cysteine protease, dessication-related protein, and two unknown protein. These proteins were involved in energy and metabolism, chaperon, antioxidation, apoptosis, which might relate to seed abortion of longan.

  • Abstract: Two different full lengths of EG cDNAs, termedas LcEG1 and LcEG2, were isolated from litchi fruit using RT-PCR and RACE (rapid amplification of cDNA ends) methods. Sequence alignment showed that four typical Cys residues and two predicted glycosyl hydrolase activesites were observed in the deduced LcEG1 and LcEG2 proteins.Northern blotting analysis showed that the level of LcEG1 mRNA accumulation gradually decreased in pericarpbut steadily increased in aril accompanying with fruit growth and development, while the expression of LcEG2 could only be detected in aril at early stage of fruit development.More importantly, the accumulation of LcEG1 mRNA was higher in cracking-susceptible cultivar Nuomici than that
    in cracking-resistant cultivar Huaizhi at any stage of fruit development.Thus, it could be speculated that LcEG1 was closely related to the growth and cracking of litchi fruit, while, LcEG2 was only related to the growth of fruit at the early phase.

  • Cloning of L-galactose Dehyrogena se Gene from Actinidia latifolia Merr. and Its Expression in E.coli

  • Abstract: Using fruits of Actinidia latifolia Merr. with the highest content of vitamin C in Actinidia as materials, the fragment of L-galactose dehydrogenase (GalDH) gene designated A lGalDH (GenBank accession No. EU525846 ) was amp lified by reverse transcrip tion polymerase chain reaction ( RT-PCR ) and then cloned. Bioinformatics analysis indicated that the length of cDNA was 997 bp, which contained an open reading frame of 960 bp and encoded a p rotein of 319 amino acid residues. The homology analysis demonstrated that A lGa lDH shared high similarities of nucleotides and deduced amino acids with those in other plants over 76% and 70% respectively. The forms of A lGalDH included one Aldo2keto reductases conversed domains.
    The A lGa lDH was then successfully subcloned into pET232a ( + ) to construct its p rokaryotic exp ression vector pET2A lGalDH. After transformation into E1coli BL21, the fusion p rotein (6 ×His2A lGalDH) was produced at a high level induced by 011 mmol·L - 1 IPTG. One single band of p rotein with the His-tagwas purified by Ni-His Bind Resin and then used as the temp late to analyze the enzyme activity which showed 120 pmol·min-1 ·mg-1.

  • Hot pepper variety‘Huaxi’was used to study the effects of Ca2 + on the growth and enzyme activities related to respiratory metabolism of hot pepper seedling root at different time (0, 3, 6, 9 and 12 d)after flood stress. The results showed that exogenous Ca2 + could significantly reduce damage under flood stress. The root growth of hot pepper could be improved, and total length, total surface area, total volume,root hair number and root diameter were significantly larger than those of flood stress. Exogenous Ca2 + increased root activity, decreased electrolyte leakage, enhanced ADH activity, reduced LDH activity, compared to those of flood stress, which avoided the accumulation of lactic acid and acetaldehyde, kep t higherMDH and
    SDH activities and a certain level of aerobic respiration. Exogenous Ca2 + might alleviate damage to the seedling root of hot pepper under flood stress by regulating respiratory metabolism.

  • The eight Cucum is germplasms were selected for evaluation of their resistance to the root-knot nematodes Meloidogyne incognita. The results showed that the two Cucumis wild relatives were highly resistant to Meloidogyne incognita, and the six varieties of cucumber were all susceptible. The correlation analysis between the rates of changes of superoxide dismutase ( SOD) , peroxidase ( POD) , phenylalanine ammonialyase ( PAL) in roots before and after inoculation and disease index for Cucumis species tested was performed.The results showed that there was a significant positive correlation of SOD ( r = 0.8278*),highly significant negative correlation of POD ( r = - 0.9679** )
    and significant negative correlation of PAL(R=-0.8499*) associated with the disease index, respectively. POD isoenzyme electrophoresis analysis showed that there were three more enzyme bands (Rf = 0.126, 0.188, 0.944) in the resistant genotype ‘C. hystrix’, but only one weak band (Rf = 0.396) in the suscep \tible genotype‘Beijing Jietou’when compared to noninoculated control. The level of SOD, POD and PAL activities can be used as the biochemical markers and the supplement for the conventional method of evaluation for resistance to the root-knot nematodes in Cucumis species.

  • The genetic effects of main traits in bitter gourd were analysed by using genotype ×environment interaction model with 7 inbreed lines and their 21 hybrids culturing in spring and autumn. The results indicated that most tested traits including early fruit yield per plant ( efy) , fruit weight ( fw) , fruit length
    ( fl) , fruit diameter ( fd) , pericarp thickness (p t) , index of fruit shape ( ifs) were mainly controlled by the additive effects, their proportion of additive variances in phenotypic effective accountive was 45.2%, 70.7%,89.4% , 38.1% , 41.2% and 22.0% respectively, all of which were significant at 0.01 level; However,fruit yield per plant ( fy) and fruit number per plant ( fn) were significantly more impacted by additive ×environment interaction than that by additive effects, the variance proportion of which were 24.5% and 38.1%.
    All the economic traits influenced by dominance or dominance ×environment, but their variance proportion was smaller compared to additive or additive ×environment. General heritabilities in the narrow and broad sense were significant at 0.01 level for all traits except fn. Interaction heritabilities in the narrow and broad sense for fn were the highest in those of the investigated traits and significant at 0.01 level.

  • Xinjiang Hami melon, with independent and different genetic background of cantaloupe, is a unique cultivar in China. A set of populations were developed from a cross between the cantaloupe [ Cucumis melo L. ssp. melo convar. cantalupa ( Pang. ) Greb. ] and the Xinjiang Hami melon [ Cucumis melo L.
    ssp. melo convar. ameri ( Pang. ) Greb. ]. Simple sequence repeat ( SSR) markers were used to construct permanent genetic map with the F2 S6 populations. The map consisted of 22 linkage groups, contained 201 SSR
    markers and 1 morphological trait, it spanned a total genetic distance of 977.153 cM with an average map interval of 4.837 cM between two markers. Seventeen linkage groups in the map were collinear with cantaloupe genetic map, 46 SSR markers were common in two maps. The construction of Xinjiang Hami melon SSR map laid foundation for the continuation of the international integration of melon common genetic map and comparative study.

  • The authors analyzed anthocyanins contained in the Senecio cruentus DC. flower with different colors. The cluster analysis made based on the content of anthocyanins in the different flower colors of S. cruentus. The floral color was measured by a spectrophotometer (NF333 ). A high performance liquid
    chromatography with a photodiode array detector (HPLC - PAD) and HPLC - electrospray ionization - mass spectrometry (HPLC - ESI - MS) methods were employed for qualitative and quantitative analysis of anthocyanins and flavonols. The major anthocyanin and the relationship between the appearance and the content of anthocyanins are found through the cluster analysis. There is a linear negative correlation between the flower lightness and the total anthocyanins contained in flower petals. The blue and red flower colors of the S.cruentus decided by delphinidin (Dp) and cyanidin (Cy). The pink flowers contained the derivatives of cyanidin (Cy) and pelargonidin ( Pg). The purple flower contained the derivatives of delphinidin and cyanidin. The lighteness of flower has a negative correlation with the quantity of anthocyanins and the level of redness has a positive correlation with Cy-type anthocyanins contained in flower. The flower color of the S. cruentus is decided by the composition and quantity of the anthocyanins.

  • The effect of DNA methylation inhibitor ( 5-azacytidine, 5-azaC) to chrysanthemum (Dendranthem a ×grandiflorum ) has been studied by the method of in vitro treatment. The results indicated that 500μmol·L - 1 of 5-azaC caused death, above 100μmol·L - 1 of 5-azaC inhibited the materials’development. The multiple buds differentiation of chrysanthemum was inhibited by every concentration of 5-azaC and the inhibiting effect of 5-azaC has time and dose additive effect. 5-azaC treatment at low concentration induced
    the flowering early, this effect were stability and could be inherited through asexual multiplication. The results of MSAP (methylation sensitive amplified polymorphism technique) indicated that, the genomic of treated materials DNA methylation decreased distinctly. Part of the sites kept demethylation status in the successive transfer culture when 5-azaC removed.

  • RcHSP17.8 is a high temperature (HT) induced expressing gene, by RT-PCR and Southern blot indicate that there are several copies of RcHSP17.8 in genome DNA of Rosa hybrida L. RcHSP17.8 is more strongly expressed in varieties resistant to HT, such as‘Schloss Mannieim’, ‘Radio’, ‘Las Vegas’,
    ‘Caprice de Meiland’, ‘Girl’, meanwhile weakly or not expressed in varieties sensitive to HT, such as‘Kordes’Perfecta’, ‘Hiogi’, ‘Pfalzer Gold’. The results indicate that this gene is possibly relative to the
    tolerance in Rosa hybrida L. to HT. In order to study its possible function, RcHSP17.8 gene has been transferred into Escherichia coliBL21. Recombinant RcHSP17.8 was overexpressed in the recombinant E.coli cells which showed imp roved viability under different abiotic stress conditions, such as, high salt concentration,heavymetal and oxidative stress, as compared with control cultures. Those resultswould provide molecular identification principle to selecting varieties of Rosa hybrida L. with strong tolerance to HT.

  • The utilization, residue, loss and transport dynamics of nitrogen on orchard via soil in early spring by seven-year-old‘Gala’apple trees (Malus domestica) / Malus hupehensis were studied, using tracertechnique of 15N. The results showed that the utilization ratio of N fertilizer was low. At full-bloom stage, the utilization ratio of N fertilizer was lower (11.38% ). After fruit harvest, the utilization ratio of N fertilizer was the highest. At full-bloom stage, the residue rate of N fertilizer was the highest ( 57.10% ). After fruit harvest, the residue rate of N fertilizer was the lowest. The residue rates of different phenophase are different. At full-bloom stage, the residue rate of N fertilizer of 20 - 40 cm layer was the highest and, after fruit harvest,it was the lowest. At the new shoot growth stage, the residue rate of N fertilizer ( 0 - 20 cm layer) was the highest and, at fruitmaturity stage, it was the lowest. However, the residue rate of N fertilizer (40 - 60 cm layer) was up to the highest at fruit rap id-swelling stage. After fruit harvest, the residue rate of N fertilizer (0- 20 cm layer) was higher than that at fruit maturity stage and the residue rate of N fertilizer (other layers)was the lowest. On the contrary, the loss rate of N fertilizer was improved gradually with the process of phenophase. At full-bloom stage, itwas the lowest to 31.53% , and after harvest, the tip top was 58.40%.

  • In vitro-grown shoot tip s of Stachys floridana Schuttl. ex Benth. were successfully cryopreserved by the encapsulation - vitrification method. Excised 5 mm shoot tip s, subcultured 4 times and domesticated at 5 ℃ for 14 d, were precultured for 1 d in liquid MS medium supplemented with 0.5 mol·L - 1 sucrose. Precultured shoot tips loaded with a mixture of improved MS and 2PVS2 for 20 min at 25 ℃ were directly dehydrated with a highly concentrated vitrification solution ( PVS2 ) for 4 h at - 20 ℃ and exchanged fresh PVS3 prior to immersion in liquid nitrogen for 24 h. After rewarming at 40 ℃ for 1 min, the cryopreserved tips were washed twice in imp roved MS liquid medium supp lemented with 1.2 mol·L - 1 sucrose for 2 ×10 min and then post-cultured on regeneration medium in dark for 20 d, the rate of root recovery amounted to about 63.7%

  • Antibacterial effects of volatile compounds from leaves of six perennial root flower p lants widely grown in Guanzhong area of Shaanxi, China, were investigated on Staphylococcus aureus ATCC29213, Escherichia coli DH10B, B acillus subtilis FM2S2 and Saccharomyces cerevisiae. Results showed that although all the tested plants exerted antibacterial effects, Dendranthema morifolium ‘ Guifeihong’, Paeonia mairei, Hemerocallis fulva‘Stella Deora’, Hemerocallis fulva‘Stella Deora’ + Paeonia mairei and Iris brevicaulis + Paenoia
    mairei were found most effective in inhibiting bacteria, with their inhibitory effects reaching higher than 30%.

  • The multiplex RT-PCR assay was established to simultaneously detect Apple stem grooving virus (ASGV) , Apple chlorotic leaf spot virus (ACLSV) and Apple stem pitting virus (ASPV). Using tissue culture apple seedling (Wangshanhong) infected by three latent apple viruses as sample, a series of factors affecting the multip lex RT-PCR were optimized. Sensibility test showed that it could simultaneously detect out ASGV, ACLSV and ASPV in 187.5 ng total RNA. The multiplex RT-PCR products were cloned and sent for sequencing, and the sequences show high homologies to the published virus sequences. Consistent results for virus detection of 12 apple samples in field were obtained by the multiplex RT-PCR and single RT-PCR system, suggesting the accuracy of the system in ASGV, ACLSV and ASPV detection.

  • During the last twenty years, DNA sequences have been widely used in phylogenetic analyses of plants at the different taxonomic levels, and provided convincing evidence to solve the long-standing disputed or unsolved phylogenetic problems. In this paper, the current status of applications of DNA fragments in plant phylogenetic studies were briefly reviewed taking Rosaceae as an examp le, and the common troubles occurring in phylogenetic studies and the possible reasons were discussed in detail. Finally, the strategies and
    points for attention in selecting DNA sequences were proposed for phylogenetic reconstruction of plant taxa especially for plants that have undergone complex evolutionary processes such as hybridization, polyploidization and rapid radiation.

  • ‘Baicui Jimali’ ( Prunus salicina‘Baicui Jimali’) is a new and good quality plum cultivar selected from ‘Sanhuali’. The fruit shape is oval. The average weight of fruit is 52.7 g. The soluble solids content is 9.5% - 11.5%. The fruit is larger and uniform. There is not residual astringency in fruit peel. The flesh is light red, sweet, tender and crisp, it is a rare white flesh cultivar in ‘Sanhuali’plum. It is fit to be planted in south subtropical mountainous regions, the fruit ripen from late June to early July in Guangdong Province.

  • ‘Sufen 8’ is a new tomato hybrid which developed by crossing‘GB9736’as a female parent with‘TM9761’as a male parent. It is indeterminate growth type, which grows vigorously. The leaf is dark green and the ripe fruit is pink. The average weight of single fruit is 220 g. It has excellent characters such as
    big fruit, regular shape, good commerciality and high productivity. It is resistant to ToMV, leaf mold and wilt, tolerant to low temperature. It is suitable for p rotected cultivation.

  • ‘Qingfeng’is a new eggplant hybrid developed by crossing two inbred lines‘Changshixuan’and‘Taixuan 6’in 2004. In South China it is suitable for cultivation in open field in spring and autumn. It produces long cylindrical fruits with purple skin, 28 - 30 cm in length and 512 cm in diameter. The average
    fruit weight is 260 g. The flesh is green-white, tender and good quality. It has middle resistance to bacterial wilt and is tolerant to heat stress.
