Studies on the Culture Solution for Lily Pollen Vitality Test

Abstract: The fresh pollen of oriental hybrids ‘Sorbonne’was emp loyed as experimental material to carry on the studies on culture solution for the determination of lily pollen viability. Comparisons were made among the effects of different concentration of sucrose, boron, calcium, magnesium and potassium on the germination of lily pollen through simp le factorial experiment. And an orthogonal design was used on that basis to compare the effects of sucrose, boron and calcium on the germination of lily pollen. Results showed that sucrose and boron have great effects on the germination of lily pollen. The op timum culture solution was sucrose 100 g·L - 1 +H3BO3 20 mg·L - 1 +CaCl2 20 - 30 mg·L - 1. When cultured in pure water, the protoplasm of the pollen would split, and the substance inside would flow out; when cultured in the solution with high concentration of sucrose or salt, the protoplasm would dehydrate and separate from the cell wall. These two would inhibit the germination of lily pollen.
