Expression Analysis of Genes Related to Photosensitive Color Under the Caylx in Eggplant Based on Transcriptome Sequencing

为了探究茄子光敏品种和非光敏品种控制萼下果色的差异基因,以萼下紫色的圆茄自交系Y5和萼下绿色的圆茄自交系Y73萼片下果皮为试材,进行去萼片见光0、4和8 h的转录组测序。共获得75. 61 Gb有效数据(Clean data),Q30碱基百分比均达到93. 97%以上;两个自交系材料在3个时间点的转录组测序比较共获得11 812个差异表达基因,包括5 277个上调基因和6 535个下调基因。3个时间点数据KEGG富集分析同时指向苯丙烷生物合成、类黄酮生物合成和光合生物的固碳作用通路。苯丙烷和类黄酮生物合成路径中共同差异表达的基因有4个,分别是FAOMTCCOAOMTCYP73A4HST。筛选出了与花青素合成相关结构基因CHIANSF3HPAL以及转录因子基因MYB113在萼下果皮表达差异显著。

In order to explore the different genes related to low light tolerance between the the photosensitive and non-photosensitive varieties,stable genetic purple under the calyx,round eggplant inbred line Y5 and green under the calyx,round eggplant inbred line Y73 were sequenced at three-time points(0,4 and 8 h). The total effective data of 75.61 Gb was obtained,and the percentage of Q30 base reached 93.97%. 11 812 differential expression genes were obtained by sequencing and comparing two inbred lines at three-time points,including 5 277 up-regulated genes and 6 535 down-regulated genes. KEGG enrichment analysis of data at three time points shows three same pathways of phenylpropane biosynthesis,flavonoids biosynthesisand carbon fixation of photosynthetic organisms. There are four genes in the biosynthesis pathway of phenylpropane and flavonoids,named FAOMTCCOAOMTCYP73A4 and HST. The CHIANSF3HPAL genes and the transcription factor MYB113 gene related to anthocyanin synthesis were screened to express significantly in the peel under the calyx.
