Distribution and Molecular Characterization of Citrus yellow vein clearing virus in Zhejiang Province

摘要: 运用RT-PCR技术对采自浙江12个柑橘产地的181份柑橘样品进行检测,首次从浙江的‘佛手’柑(Citrus bergamia)和‘红美人’橘(C. reticulata)上检测出柑橘黄脉病毒(Citrus yellow vein clearing virus,CYVCV),其检出率分别为68.0%和47.2%。选取4个CYVCV毒株与11个已知CYVCV毒株进行全序列分析,结果显示CYVCV序列保守性较高,所有15个CYVCV毒株的核苷酸和氨基酸序列相似性分别为96.9% ~ 99.8%和97.9% ~ 99.4%。虽然采自浙江的4个CYVCV毒株在进化树中聚成单独的簇,但CYVCV毒株间的亲缘关系可能不与其采样地的存在相关性。

Abstract: In this study,181 citrus samples collected from 12 districts and counties in Zhejiang Province were detected by reverse transcription-polymerase chain reaction(RT-PCR). The results showed that Citrus yellow vein clearing virus(CYVCV)was first detected in Bergamot(Citrus bergamia)and Ehime Kashi 38(C. reticulata)from Zhejiang Province with an incidence of 68.0% and 47.2%,respectively. The complete genome sequences of 4 CYVCV isolates from Zhejiang Province were sequenced and characterized. Comparisons of the whole genome sequences of these 4 CYVCV isolates as well as 11 isolates previously reported from around the world revealed the sequence identity ranged from 96.9%–99.8% at nucleotide level and 97.9%–99.4% at amino acid level,respectively,indicating that there is a very low level of sequence heterogeneity among CYVCV isolates of different geographic origins and hosts.Although CYVCV isolates from Zhejiang Province were clustered in the same clade in the phylogenetic tree,other isolates from China did not show a relationship with the geographic origin of the sampled trees.
