Cloning and Expression Analysis of Lectin Genes from Pleurotus ostreatus

Abstract:

Two lectin genes(Plectin1 and Plectin2from Pleurotus ostreatus have been cloned based on rapid amplification of cDNA ends(RACE) method in this study. The full-lengths DNA of Plectin1 and Plectin2 are 1 371 bp,1 359 bp,and contain 5 exons and 4 introns,respectively. The open reading frame(ORF)of Plectin1 is 1 134 bp encoding 377 amino acids,whilethat of Plectin2 is 1 122 bp encoding 373 amino acids. Southern blot confirms both Plectin1 and Plectin2 had only one copy in the genome of P. ostreatus. qRT-PCR analysis shows Plectin1 and Plectin2 were highly expressed in mature fruiting body stage and young fruiting body stage respectively. This suggests Plectin1 and Plectin2 may play an important role in P. ostreatus fruiting body development.
