辣椒转录组SSR挖掘及其多态性分析

摘要: 利用SSR检索程序从两个辣椒转录组共221 037条unigenes(97.3 Mb)中筛选得到17 319个SSR位点(7.83%),其发生频率为1/56 kb。其中以单核苷重复基序为主导类型,占总SSR的56.30%;其次是二、三核苷酸重复基序,其出现频率分别为19.16%和23.18%。对所有含SSR位点EST序列设计的10 468对引物进行E-PCR多态性检测,获得1 538对E-PCR多态性引物。随机选取20对引物进行验证,其中7对(36.84%)在10个不同类型辣椒材料中表现出多态性,PIC值范围为0.33 ~ 0.89,平均为0.67,将10个辣椒材料分为3类。这些潜在的多态性EST-SSR的开发为辣椒遗传多样性分析、图谱构建提供了更丰富的候选分子标记。

Abstract: In this study,we investigated 221 037 unigenes(97.3 Mb)from two transcriptome in pepper using SSR finding soft. In total,17 319(7.83%)SSRs were identified from these SSR-containing unique ESTs with the occurring frequency of 1/56 kb. Among,single nucleotide type(56.30%)is the most abundant,followed by di-(19.16%),tri-(23.18%). A total of 10 468 primer pairs were successfully designed and 1 538 primer pairs exhibited polymorphism by E-PCR 19 out of 20 primer pairs selected at random yielded amplification products,of which 7 primer pairs showed polymorphism among 10 different pepper varieties,PIC ranged from 0.33–0.89,average 0.67,and 10 pepper breeds were grouped into three classes. This results indicate that molecular markers will be more usable in map structure,analysis of genetic polymorphism in pepper.
