Effects of GMPase Overexpression on Ascorbic Acid Content and Relative Index to Low-temperature Tolerance in Tomato Plants

摘要: 采用农杆菌介导法获得了GDP–D–甘露糖焦磷酸化酶基因(GMPase)超表达的转基因番茄植株,并且采用Western blot检测到有两株GMPase蛋白表达显著增加。比较了这两个株系和野生型植株中抗坏血酸含量及耐低温能力。与野生型番茄相比,GMPase超表达在常温及低温胁迫下均使GMPase活性提高,使AsA和DHA含量显著增加,降低了低温胁迫下膜脂过氧化产物MDA的积累。

Abstract: Two transgenic tomato plants of overexpressing GDP-D-mannose pyrophosphorylase gene (GMPase)were obtained by Agrobacterium-mediated transformation,and Western blot analysis indicated that the GMPase protein in transgenic plants was significantly increased. T0 make sure the function of GMPase overexpression,ascorbate content and cold tolerance capacity were studied. Compared with wild-type plants,the GMPase activity and contents of AsA and DHA in both normal and low-temperature conditions were increased and the MDA content was decreased in the transgenic plants. Based on these results,it can be concluded that the overexpression of GMPase may increase tomato tolerance to cold stress by increasing AsA level and lowering lipid peroxidation.
