Investigation and Analysis on the Genetic Diversity of Persimmon Germplasms in Plateau of Northwest Guangxi

Abstract: The germplasm resources of persimmon in Yungui plateau of northwest Guangxi were investigated and researched. The collected germplams which included original wild type were analyzed by morphology and SCoT molecular marker. The results showed that Yungui plateau of northwest Guangxi possessed rich persimmon germplasm resources,31 different accessions of germplasm which belonged to three species Diospyros kaki Thunb.,D. oleifera Cheng and D. kaki var. silverstris Mak. were corrected. The corrected persimmon germplasms had large variations on morphology as well as at the level of DNA. Thirty-five independence morphological characters were used to conduct the Q-style systematic clustering analysis and the 31 accessions of persimmon germplasm were divided into 5 groups at the Euclidean distance of 9.8,which accorded with their fruit shapes and germplasm types. Thirteen primers were used to amplify the persimmon genome DNAs and 161 SCoT loci were produced,154 of which were polymorphic loci and with a polymorphism rate of 95.65%. The cluster analysis conducted by UPGMA showed that 31 accessions of persimmon germplasm could be distinguished from each other and were divided into five groups at the similarity coefficient of 0.686,which was similar to the result of morphology analysis.
