Cloning and Sequence Analysis of Chalcone Synthase Gene in Ornamental Peach

Abstract: The Chalcone Synthase(CHS)coding sequence(CDS)was cloned from the ornamental peach‘Hongyu Chuizhi’by using SON-PCR techiniques. It was designated as PphCHS,and the GenBank accession number was JN391444. The full CDS was 2 353 bp in length with an open reading frame(ORF)of 1140 bp,encoding a protein of 379 amino acids. Homology analysis of the deduced amino acids indicated that the sequences had more than 90% similarity with those of other reported plants. Amino acids cluster analysis showed that CHS from Prunus cerasifera was clustered together with PphCHS firstly,followed by that from strawberry. Bioinformatic analysis results indicated that the PphCHS amino acids had no signal peptide sequences,but possessed the CHS family’s characteristic sequences,RLMMYQQG CFAGGTVLR.
