A Study on Obtaining Primary Trisomics by the Isolated Microspore Cultureof Autotetraploid Chinese Cabbage

Abstract: Selection of primary trisomics of Chinese cabbage 〔Brassica campestris L. ssp. pekinesis (Lour) Olsson〕is an important foundation for the gene chromosome mapp ing and other genetic studies. The aneuploid plants with one extra chromosome (2n + 1) were obtained from the isolatedmicrospore cultured plantlets of the tetraploid Chinese cabbage. By karyotype analysis of the root tip metaphase chromosome, meiosis observation and plant characters investigation, a set of primary trisomics ( Tri-1, Tri-2, Tri-3, Tri-4, Tri-5, Tri-6, Tri-7, Tri-8, Tri-9 and Tri-10) was identified. Each trisomic was with some unique characters on plant morphological traits. The isolated microspore culture of tetraploid Chinese cabbage is a convenient and effective way to select trisomics in Chinese cabbage.
