Isolated-m icrospore Culture and Plantlet Regeneration in Cabbage ( Brassicaoleracea L. var. capitata L. )

Abstract: Some factors affecting isolated2microspore culture of cabbage (Brassica oleracea L. var. capitata L. ) were studied using double-crossed and tetra-crossed hybrids as the donor plants. Results showed that the genotype was one of the most critical factors affecting embryogenesis in isolated microspore culture. The embryogenesis capacity of the low responsive genotype could be significantly improved after crossed with the genotype with high embryogenesis ability. The tetra-crossed hybrid could produce more vigorous embryos and result higher seedling rate compared with the double-crossed hybrid. The yield of embryoid could be significantly increased by adding new liquid medium with lower sucrose concentration into the original culture medium with a higher sucrose concentration after 4 days of culture.
