Efficient Culture and Plan t Regeneration from Cotyledon Protoplasts of ‘Cuixiu’Cucmnber

Abstract: Isolated and purified cotyledon protoplasts of cucumber(Cucummis sativust L. ‘Cuixiu’)were cultured at diferent population density ranging from 5.5×10/mL to 5.5×105 /mL in mKM8p liquid medium respectively an d underwent cell division in varying degrees.Sustained cel divisions with eventual somatic colony production occurred only when the population density of protoplasts exceeded 1.0×103/mL. the highest plating efficiency was about 54% .Lager quantifies of embryoids diferentiated from protoplast derived ealli and then converted into planflets after succedent induction culture.Results demonstrate that the concentration of Ca2+ has an important effect on the stability an d cel division of cucumber protoplasts.
