白菜病毒诱导基因沉默技术体系的建立

摘要: 以白菜(Brassica campestris ssp. chinensis Makino)品种‘苏州青’为材料,利用芜菁黄化花叶病毒(Turnip yellow mosaic virus,TYMV)质粒pTY成功抑制了白菜叶片八氢番茄红素脱氢酶(phytoene desaturase,PDS)基因的表达,验证了病毒载体pTY在白菜上诱导基因沉默的能力,建立了白菜病毒诱导基因沉默VIGS体系。为了进一步验证病毒载体pTY在白菜上诱导基因沉默的能力,以BcNRT1为靶基因,构建pTY-BcNRT1质粒侵染白菜后,显著抑制了BcNRT1在转录水平的表达。

Abstract: In this study,a viral vector derived from Turnip yellow mosaic virus(TYMV)was used to induce virus-induced gene silencing(VIGS)of phytoene desaturase gene(PDS) in non-heading Chinese cabbage(Brassica campestris ssp. chinensis Makino.)cultivar‘Suzhouqing’and the result showed that the expression of PDS was successfully repressed. The ability of inducing silence was confirmed and the VIGS system was established. To further confirm the VIGS effect on non-heading Chinese cabbage,BcNRT1 was selected as the target gene,and the expression of BcNRT1 was successfully inhibited after inoculated.
