月季bHLH基因的克隆、表达及其与MYB和WD40的互作分析

摘要: 以月季‘月月红’(Rosa chinensis‘Slater’s Crimson China’)花瓣为材料,利用同源克隆法鉴定到1个RcbHLH基因的cDNA全长序列,并研究了其生物信息学特征、组织特异性表达及其与MYB和WD40的互作关系。序列分析表明,RcbHLH的开放阅读框长2 112 bp,编码703个氨基酸,基因登录号为KY783912。结构域序列分析显示,RcbHLH为一个保守的N端含有碱性氨基酸的DNA结合区、C端含有α螺旋–环–α螺旋的bHLH蛋白。对RcbHLH进行系统进化分析发现,RcbHLH与草莓及其他蔷薇科植物的bHLH蛋白具有较高的同源性。组织特异性实时定量RT-PCR分析揭示RcbHLH基因主要在花瓣中表达。酵母双杂交结果显示,RcbHLH能与PavMYB和PavWD40相互作用形成MYB-bHLH-WD40复合物。以上结果为进一步研究RcbHLH的功能鉴定奠定了基础。

Abstract: The complete cDNA sequence of RcbHLH gene was identified to reveal the underlying anthocyanin synthesis mechanism by homology-based cloning in the‘Slater’s Crimson China’(Rosa chinensis). The bioinformatic characteristics,tissue specific expression pattern,and interaction of this gene with MYB and WD40 were investigated. These results showed full length open reading frame of RcbHLH gene was 2 112 bp. It encoded 703 amino acids(Accession number:KY783912). The RcbHLH had a conserved base domain in its N terminal and a conserved Helix-Loop-Helix domain in its C terminal. The results of analyzing protein system evolution showed that RcbHLH had high homology with the strawberry bHLH and other bHLH protein of plants in Rosaceae. qRT-PCR showed RcbHLH gene was mainly expressed in petal. The results of yeast two-hybrid experiments indicated a MYB-bHLH-WD40 (MBW)complex was formed through the interacting of RcbHLH,PavMYB and PavWD40. These results provided basic information to further illuminate the functions of the RcbHLH gene.
