芥菜型油菜雄性不育系与甘蓝远缘杂交胚培养及早代育性鉴定

摘要: 为了将优异的芥菜型油菜雄性不育性转育到甘蓝中,对甘蓝和芥菜型油菜不育系远缘杂交胚的发育情况进行了研究,结果发现杂交胚发育10 d左右后开始败育。在杂交胚发生败育之前进行胚离体培养,在MS基本培养基中添加1 mg · L-1 BA,0.1 mg · L-1 NAA和活性炭的条件下,得到了远缘杂交胚发育而成的植株。进而利用石蜡切片对远缘杂交植株的不育性进行了显微观察,发现其败育的时期发生在雄蕊原基分化时期。

Abstract: The development of the distant hybrid embryos derived from Brassica oleracea var. capitata L. and male sterile line of B. juncea were investigated in this study. The results showed that the hybrid embryos began to abort in approximate 10 days after pollination. Some hybrid plantlets were obtained by using ovule rescue culture in vitro in MS media supplemented with 1 mg · L-1 BA,0.1 mg · L-1 NAA and activated charcoal. The development of anther in distant hybrid plants were examined using paraffin section,and the abortion was found in the differentiation stage of stamen primordium.
