现代月季(Rosa hybrida)叶片植株再生体系的建立

摘要: 本试验对冰山、金秀娃、火球等24个现代月季(Rosa hybrida)品种再生体系进行了研究,
结果表明:部分品种可从叶柄砧处直接再生出小苗;基因型、植物生长调节剂组合、暗培养时间及叶片的
幼嫩程度对现代月季叶片的再生影响很大,暗培养和幼叶均有利于再生。24个现代月季品种中l2个品种
再生出了不定芽;现代月季品种冰山幼叶暗培养30 d后的再生率可从无暗培养的15.6%提高到45.6% ;冰
山顶生3片叶、中部叶片及基部叶片的再生率分别为46.5% 、12.9% 和0。适宜现代月季叶片不定芽再生
的培养基为Ms+6一BA 6.0 mg·L +ZT 6.0 mg·L +IBA 0.4 mg·L~;芽伸长培养基为MS+6一BA 2.0
mg·L +IBA 0.1 mg·L~;生根培养基为l/2 MS+NAA 0.2 mg·L +0.1%活性炭。

Abstract: The regeneration system of 24 rose cultivars(Rosa hybrida)was researched.The research
results indicted that genotypes.hormone varieties and its concentrations.cultural time in the dark and ages of
the explants have great effect on the regeneration of rose leaf explants、Culturing in dark and using tender ex—
plants can improve the shoot regeneration of leaf explants.Only l 2 varieties can forlTl adventitious buds from
24 varieties of roses researched. After 30 days of dark culturing,the regeneration rate of Iceberg improved
from 15.6% to 45.6% .In vitro the regeneration rate of Iceberg,if leaves from upside of rose。iS 46.5% :
from medium is 12.9% :from downside is 0、The appropriate media for regeneration from leaf explants of rose
is MS+6.BA 6、0 mg·L一 +ZT 6.0 mg ·L一 +IBA 0.4 mg·L一 .The medium for shoot buds elongation iS
MS+6一BA 2.0 mg·L一 +IBA 0.1 mg·L一 .The medium for regenerating adventitious roots iS 1/2 MS+
NAA 0.2 mg·L +0.1% activate carbon.
