摘要: 为获得与紫薇(Lagerstroemia indica)紫(红)色叶片关联的分子标记,以19个紫薇品种及其杂交F1子代为试材,对幼叶期、完全展叶期、盛花期和落叶期的叶色进行目测和CIE L*a*b*检测。通过结合转录组和基因组的数据以及靶向测序,鉴定参与叶色形成的候选SNP位点,通过高分辨率溶解曲线(high resolution melting,HRM)对候选的SNP在F1子代中进行了检测。结果显示,目测和CIEL*a*b*均能较好地区分叶片颜色。红度a*可区分绿色叶和彩色叶(红色和紫红色),亮度值L*、黄度值b*以及彩度值C和色调角度h在三种叶色(绿色、红色和紫红色)中均有显著差异。本研究中共筛选获得53个候选SNP位点,靶向测序结果显示6个位点∆SNP大于0.2。Sanger测序以及HRM分析‘红火球’和‘白云映霞’F1代的基因型结果显示,其中4个SNP与叶色相关系数介于0.184 ≤ r ≤ 0.623,达到显著水平,1个SNP标记在绿色和彩色叶片中呈现有、无多态性。紫(红)色(P)型基因型频率越高,叶片颜色越深,说明紫(红)色叶片是由多基因控制的加性效应导致。
