黄瓜未授粉子房离体培养获得胚囊再生植株

Embryonary Sac Regenerated Plants Were Obtained in Cucumber via in Vitro Unpollinated Ovary Culture

摘要: 7 种 F1 代黄瓜(Cucumis sativus L.)未授粉子房为试材进行再生植株培养,通过石蜡切片观察、激素含量测定探究胚囊再生植株形成过程及相关生理变化。结果表明:在 MS + 0.06 mg · L-1 TDZ培养基中有 3 个供体黄瓜材料未授粉子房获得再生植株,经流式细胞检测、染色体计数、SSR 验证分别为单倍体、双单倍体和同源四倍体。MS + 0.04 mg · L-1 TDZ + 1.0 mg · L-1 6-BA + 0.1 mg · L-1 NAA + 12 mg · L-1 AgNO3 培养基中子房产生大量胚状结构,但分化培养后没有获得再生植株。石蜡切片观察到在MS 和 MS + 0.06 mg · L-1 TDZ 培养基中培养 5 d 和 17 d 时胚囊已经发育。供体黄瓜子房切片中 GA、IAA含量随培养天数增加呈现先降低后升高的趋势,在培养 8 d 时最低,17 d 时 GA 含量比 2 d 时更高,IAA含量无明显变化。

Abstract: Seven kinds of F1 generation cucumber unpollinated ovary were used as test materials for regenerated plant cultivation. Paraffin sections of ovary were observed,and hormone content of ovary slices at different culture stages were measured to explore the formation process and related physiological changes of embryonary sac regenerated plants. The results showed that the regenerated plants were obtained from unpollinated ovary culture of cucumbers with three donor materials under MS + 0.06mg · L-1 TDZ medium,which were identified as haploid,double haploid and autotetraploid respectively by flow cytometry,chromosome count and SSR. A large number of embryonary like structures were produced in ovary under MS + 0.04 mg · L-1 TDZ + 1.0 mg · L-1 6-BA + 0.1 mg · L-1 NAA + 12 mg · L-1 AgNO3medium,but no regenerated plants were obtained after differentiation culture. The embryonary sac underMS and MS + 0.06 mg · L-1 TDZ medium was observed to develop on the 5th and 17th day by paraffin sections. Hormone determination suggested that the content of GA and IAA are on the decline to the lowest at the 8th day and increased subsequently. The GA content in the ovary slices at 17 d was higher than that in the culture for 2 d,but the variation of IAA content is slight.
