A Highly Eff icient System Establishmen t of Shoot Regeneration from LeafExplants of Strawberry

Abstract: An efficient and reliable method for shoot regeneration from leaf disks of strawberry (Fragaria ananassa Duch ‘Toyonoka’) was established. The optimum medium for shoot regeneration was MS supplemented with thidiazuron ( TDZ) 1.5 mg·L - 1 and Indole-3-butyric acid ( IBA) 0-4 mg·L - 1. Different color films had great effect on the shoot regeneration of strawberry. The presence of red and green films induced the maximum percentage of shoot regeneration ( > 95% ) and the highest number of buds per explant( > 25). However, the blue and yellow films inhibited the shoot regeneration of strawberry. The difference of films spectrum was focused on 300 - 700 nm. The light intensity of red and green films was weaker than yellow, blue and white ones. Dark treatment could effectively p romote shoot regeneration rate. In contrast, maximum percentage of shoot regeneration (100% ) and the highest number of buds per explant (9.05) were observed with dark treatment of four weeks.
