Cloning and Functional Research of an AGL6 Homologous Gene in Hybird of Cymbidium goeringii and Cymbidium hybirdium

Abstract: Full-length cDNA of CgAGL6(GenBank accession number GQ265900)was cloned from the floral bud of hybird of Cymbidium goeringii and C. hybirdium by rapid amplification of cDNA ends(RACE)technology. The cDNA sequence harbored an open reading frame(ORF)encoding 242 amino acids. Bioinformatics analysis indicated that CgAGL6 has two conserved domains as other AGL6 proteins:MADS-box and K-box. CgAGL6 are similar to GenBank accession L46397(Zea mays),U78782(Oryza sativa)and M55554(Arabidopsis thaliana)with homology of 70%,70% and 61% respectively. Transgenic tobacco plants ectopically expressing CgAGL6 exhibited novel phenotypes with plant size reduced significantly,flowering early,widely homeotic conversion of stamens into petal-like structures,hypogenesis of pollen,number addition or reduction of petals. These results indicated that CgAGL6 was involved in the regulation of flower transition and flower organ development.
