DAS-ELISA and Electron Microscopic Diagnosis of Tospoviruses Infecting Hymenocallis littoraris and Hippeastrum vittatum in Kunming

Abstract: The leaves of Hymenocallis littoraris(Jacq.)Salisb. and Hippeastrum vittatum(L’Hér.)Herb.,showing concentric chlorotic ring spot and brown lethal spot symptoms,were collected in Kunming. To confirm the presence of virus,the SGJ-KM and ZDH-KM infected leaves of H. littoraris and H. vittatum were detected by electron microscope observation and DAS-ELISA. Virions with a diameter of about 80–85 nm,similar in size to Tospovirus particles,were observed in the crude extracts and ultrathin sections of diseased leaves of H. littoraris and H. vittatum. The sap of diseased leaves had a positive reaction to the compound antibody of Watermelon silver mottle virus,WSMoV / Groundnut bud necrosis virus,GBNV. These results confirmed that H. littoraris and H. vittatum were infected by Tospoviruses.
