Cloning and Expression Analysis of a Floral Organ Identity Gene ClAP1 from Prunus lannesiana

Abstract: MADS-box genes encode transcriptional regulators that are critical for flowering and floral organogenesis. To find genes involved in the floral organogenesis of cherry blossom(Prunus lannesiana),cDNAs for one MADS- box genes,ClAP1,was cloned using homologous cloning and 3′ -RACE method based on the sequence database of NCBI. The full length of ClAP1 cDNA is 1 005 bp,containing an open reading frame(ORF)of 717 bp and coding for a polypeptide of 238 amino acid residues. Subsequently, sequence comparison and phylogenetic analysis revealed that ClAP1 was highly homologous to protein AP1 in Arabidopsis and belonged to the euAP1 clade of class A. However,semiquantitative RT-PCR analysis showed that ClAP1 was expressed in sepal, petal, stamen and carpel of three cherry blossom cultivars‘Makino’,‘Hisakura’and‘Albo-rosea’,which was slightly different from the expression pattern of AP1.
