Establishment of Efficient Shoot Regeneration System of Chinese Cabbage (Brassica campestris ssp. pekinensis) Inbred Line‘AB-81’and Studies of Transient Expression of gus A Gene

Abstract: An efficient shoot regeneration systemwas established using cotyledon and leave cuts of the inbred line‘AB-81’as explants. On the MS medium supplemented with 10 mg/L AgNO3 , 0. 2 mg/L TDZ, 2 mg/L NAA and 0.25mg/L ABA , the regeneration frequency of the cotyledon and leave explants was up to 93. 3% and 90% respectively. The mean number and the most number of shoots per explant was 3 - 6 and 21 respectively. Cotyledons of AB281 were inoculated with Agrobacterium tumefaciens strain EHA105 containing pMOG410 carrying gus A-intron gene. The optimum conditions for the transient expression of gus A gene were as follows : preculture time of 2 - 3 days , bacterium density of OD 0. 3 - 0.5 , infection time of 2 - 10 min , and coculture time of 2 - 3 days.
