深色有隔内生真菌对枸杞根腐病菌抑菌活性

Study on Antibacterial Activity of Dark Septate Endophytes Against Fusarium oxysporum in Chinese Wolfberry

摘要:

通过组织分离法从‘宁杞5号’枸杞健康根筛选出深色有隔内生真菌(DSE)菌株,采用两点对峙法、菌丝生长速率法、孢子萌发法和盆栽试验探究其对枸杞根腐病菌(Fusarium oxysporum)的抑菌活性,为枸杞根腐病的生物防治提供潜在的资源菌。结果表明,8株DSE菌株均可定殖于枸杞根部,形成典型的DSE结构,且对枸杞根腐病菌有明显的抑制作用;其中菌株S12(Cladosporium cladosporioides)对病菌菌丝生长的抑制率最高,为81.47%;8株DSE菌株发酵滤液和挥发物质对枸杞根腐病菌的生长均有一定抑制作用,其中菌株S12发酵滤液浓度为10%时对病菌菌丝生长的抑制率最高,达78.43%;菌株M1(Alternaria alternata)发酵滤液浓度为50%时对病菌分生孢子萌发抑制率最高,为91.48%;菌株M1产生的挥发物质对病菌菌丝生长的抑制率最高,达94.10%。盆栽试验中,菌株M1和S12均表现出较好的生防效果,防效分别达89.54%和83.17%,优于50%多菌灵湿性粉剂500倍液处理。本结果表明DSE菌株M1和S12可有效防治枸杞根腐病,具有良好的生防潜力和应用前景。

Abstract:

By tissue isolation method of the local isolate of Dark septate endophytes(DSE)strains from healthy roots of Lycium barbarum‘Ningqi 5’in order to further provide potential resources bacteria for the biological control of Fusarium oxysporum. The confrontation method,mycelium growth rate method,spore germination method,and pot experiments were conducted to evaluate the antibacterial activities of DSE against F. oxysporum in L. barbarum. The results showed that eight DSE strains could effectively colonize the roots of L. barbarum and form a typical DSE structure. In the meanwhile,all eight DES strains showed a significant inhibitory effect against F. oxysporum,the Cladosporium cladosporioides strain S12 had the highest inhibition rate of 81.47% on the mycelial growth of F. oxysporum;The fermentation filtrate and volatile substances of the eight DSE strains exhibited certain inhibitory effects on the growth of F. oxysporum,and the fermentation filtrate of strain S12 with a concentration of 10% showed the highest inhibitory effect on F. oxysporum mycelial growth,reached up to 78.43%;The fermentation filtrate of the Alternaria alternata strain M1 with a concentration of 50% showed the highest inhibition of conidial germination with 91.48%. Meanwhile,the volatile material produced by strain M1 exhibited the highest inhibition of mycelial growth with 94.10%. In the greenhouse pot experiment,inoculation with strains M1 and S12 provided a good biological control effect,with efficacy up to 89.54% and 83.17%,respectively,better than a treatment of 500× dilution 50% carbendazim wettable powder. These results demonstrate that DSE strains M1 and S12 can effectively control the root rot of disease L. barbarum,while having good potential for biological control and application.
