The Development of Multiplex EST-SSR Markers to Identification ChineseCabbage [ Brassica campestris L. chinensis ( L. ) Makino and Brassicacampestris L. pekinensis (Lour. ) Olsson] Cultivars

Abstract: In order to identify Chinese cabbage [Brassica campestris L. chinensis (L. ) Makino ] and [Brassica campestris L. pekinensis (Lour. ) Olsson ] cultivars, six multiplex EST-SSR sets made up of three pairs of primers in each setwere developed and optimized PCR condition for each multip lex SSRs amplifying.
These sixmultip lex EST-SSR markersmay distinguish the forty-three Chinese cabbage cultivars completely and test the uniformity within the individuals of three Chinese cabbage hybrids. Efficiency from single primer pair and multiplex primer sets were compared. They were measured the extent of diversity between the forty-three Chinese cabbage cultivars, uniformity within cultivars and stabilities, repetition. According to the cultivar and the locus examined. These results validated that the sixmultip lex EST-SSR markers could be used to identification Chinese cabbage cultivars and test the purity of hybrids
