Studies on Optimal Detection System of Immunofluorescence Labeling for Cell Microtubules in Brassica campestris ssp. chinensis

Abstract: In this study orthogonal design was used to optimize the system of immunofluorescence labeling for microtubules in Brassica campestris ssp. chinensis in four factors (enzyme concentration, digesting time, dilution of antibodyⅠ, dilution of antibodyⅡ)at three levels respectively. A suitable detection system of immunofluorescence labeling for cell microtubules was established: enzyme (pectinase and cellulose) concentration was 1.0%, digesting time was 1 h; antibodyⅠ was diluted 1:100 for 1 h; antibodyⅡ was diluted 1:500 for 1 h. The microtubules could be observed effectively in Brassica campestris ssp. chinensis with this system.
