Callus Induction and Plant Regenera tion from Stem Segment of Bougainvilleaglabra Choisy

Abstract: This papermainly dealt with the studies on callus induction and plant regeneration from stem segment of Bougainvillea glabra Choisy. The results showed that MS medium supplemented with BA 3.0 mg·L-1 + 2,4-D 0.2 mg· L - 1 +NAA 0.1 mg·L - 1 was best for callus induction in which the induction rate and differentiation rate of callus were 78.2% and 25.6% , respectively. Two types of shoots were observed: the first type of shootswas directly induced from the bottom of stem segment, the second type of shoots differentiated from the callus induced at the bottom of stem segment. MS medium containing BA 2.0 mg·L-1 +NAA 0.1 mg·L-1 was best for clump shoots induction in which the ratio of multiplication was 5.4, while BA and 2,4-D increased to 3.0 mg·L-1 and 0.5 - 1.0 mg·L - 1 respectively would result in leaf variation of regenerated plants. MS medium containing BA 0.2 mg·L - 1 + GA3 0.5 mg·L-1 +NAA 0.1 mg·L - 1 gave best result for efficient seedling culture. MS medium containing NAA 3.0 mg·L - 1 was best for root induction where the rate of rooting was 88.3%.
