Abstract: Preliminary Study on Tissue Culture of Bermudagrass and Its Dwarf Sonmclonal rianth Shaoy, Guo Zhenfei,and Chen Yongchuan(Laboratoryfor Ttwfgrass Science,College of Science,5China,510642,China)The stolonous nodal segments of hybrid bermudagrass cuhivar‘Tifeagle’(Cydon dtylon×C.tranmxta&nsis)were sterilized by 0.1% HgC12 and placed on solid medium contained N6 macronutrient elements,B5 micronutrient elements, MS ferric salt, Gamborg’S vitamin and additives including 2 mg.L 2,4D and 0.02 mg.L 6一BA for induction of callus.The compact embryogenic calli were obtained an d were highly regenerable after they were transferred to regeneration medium.Somaclonal variation was observed among the plantlets.One of them, T、,4, showed dwarfism variation. Compared to its parent ‘Tifeagle’, T、 showed significant morphological variation with decrease of 47% in stolon intemode length, increase of 30% in blade width ,and reduction of 32% in blad e length.TV4 had extensive root system with increase of 37% in dry weight of the newly developed adventitious roots after the stolon was planted for 40 days.TV4 showed higher chilling tolemace than ‘Tifeagle’with lower relative electrolyte leakage after the plants were subjected to 8℃ .SOD activity decreased in ‘Tifeagle’,while that increased in TV4 after chilling treatment.Th e activities of CAT an d POD dechned in both TV4 and ‘Tifeagle’.but they Were maintained at higher level in Tv4 than ‘Tifeagle’.
