4x菜薹与4x芥蓝种间杂交获得异源四倍体及其鉴定

Obtaining and Identification of the Allotetraploid from 4x Flowering Chinese Cabbage × 4x Kailan

摘要: 以四倍体菜薹(2n=4x=40)为母本与四倍体芥蓝(2n=4x=36)进行种间杂交,通过子房培养,获得了菜薹与芥蓝的异源四倍体种间杂种。研究表明,适宜菜薹与芥蓝种间杂交子房培养的培养基为Nitsch培养基,附加蔗糖70 g/L,琼脂7 g/L,维生素B1 0.5 mg/L,维生素B6 0.5mg/L,生物素0.01 mg/L,6-BA 0.5 mg/L,IAA 0.1 mg/L,pH 5.8;适宜培养的取材时间为授粉后7 d;细胞学鉴定和流式细胞仪分析表明,该杂种的染色体数为2n=4x=38,DNA相对含量约为其二倍体亲本的2倍;该杂种的开花期,叶形、叶色、花冠大小和花色等特征介于两个亲本之间。

Abstract: The allotetraploid interspecific hybrid of 4x flowering Chinese cabbage(2n=4x=40)× 4x kailan (2n=4x=36) was obtained by ovary culture. The results showed that the most suitable medium for ovary culture of 4x flowering Chinese cabbage × 4x kailan was Nitsch medium supplemented with sucrose 70 g/L, agar 7 g/L, vitamin B1 0.5 mg/L, vitamin B6 0.5 mg/L, biotin 0.01 mg/L, 6-BA 0.5 mg/L, IAA 0.1 mg/L, pH 5.8, and the best culture stage was 7 d after pollination. The cytological identification showed that chromosome number of the allotetraploid was 2n=4x=38. The amount of DNA by flow cytometry showed that DNA relative content of the allotetraploid was twice of that of its diploid presents. The morphlogical observation showed that the characters, such as anthesis, leaf shape and colour, flower size and colour, of the allotetraploid hybrid were between of its parent species.
