摘要: 以高效抗缺铁苹果种小金海棠(Malus xiaojinensis Cheng et Jiang)为试材,以异源的小麦
Nramp基因为探针进行杂交分析。Southem杂交结果表明:小金海棠基因组中存在该家族基因。Northern杂交结果表明:在根和叶中,Nramp基因在正常供铁和缺铁胁迫下均能得以转录,且叶中的转录受缺铁胁迫的诱导而加强。通过PCR方法已从基因组DNA中扩增出了752 bp的特异性产物,该片段与水稻的OsNramp3具有83% 的氨基酸序列同源性。

Abstract: To initially study the role of Nramp gene in resisfng iron deficiency stress,we analyzed this gene in Malus xiaojinensis Cheng et Jiang by using blot hybridization technique with Nramp gene fragment of wheat as probe.Southern blot result suggested that there was Nramp gene in the genome of Malus xiaojinensis.Northern blot confirmed that transcription ofthe Nramp gene is induced in both roots and leaves and strengthened in leaves by iron stress.A 752 bp fragment of Nramp gene was obtained by PCR from the genomic DNA,
which has 83% identities to OsNramp3 of rice in amino acid leve1.The results lead to the prediction that Nramp gene of Malus xiaojinensis may play a crucial role in resisting iron stress.
