绿针假单胞菌HL5-4对番茄灰霉菌的抑制活性及其定殖能力

Antifungal Activity of Pseudomonas choloeaphtis HL5-4 Against Tomato Gray Mold and Its Colonization Ability

摘要:

为探讨绿针假单胞菌HL5-4对番茄灰霉病的生防潜力,通过对峙培养和孢子萌发试验测定其对灰葡萄孢菌的抑制活性;进行离体试验和苗期试验测定其对番茄灰霉病的防治效果;采用平板稀释法测定其在番茄果实和叶片的定殖能力;并根据形态特征、生理生化特性及16S rDNA序列同源性对其进行鉴定。结果表明:HL5-4显著抑制灰葡萄孢菌的菌丝生长和分生孢子萌发,萌发抑制率为70.74%;在离体试验中,对番茄果实和叶片灰霉病的防治效果分别达到72.36%59.85%,在盆栽试验中,对番茄苗期灰霉病的防治效果为62.88%HL5-4在番茄果实和叶片保持一定的定殖密度,处理7 d时能保持104103 cfu · g-1以上;HL5-4菌株鉴定为绿针假单胞菌(Pseudomonas choloeaphtis)。

Abstract:

A promising biocontrol agent HL5-4a strain with strong effect on tomato gray moldwhich selected from the rhizosphere of healthy tea plant. In vitro and in vivo experiments were used to determine its antifungal effect and colonization ability. The results showed thatHL5-4 inhibited mycelial growth and conidia germination of Botrytis cinerea in vitro experimentthe inhibition zone of the mycelial growth was 15 mmthe inhibition rate of conidia germination was 70.74%HL5-4 performed good control potential against tomato gray moldthe control efficiencies were 72.36% and 59.85% respectively on fruits and leaves in vitro experimentand 62.88% on seedlings in potHL5-4 persisted on fruits and leaves of tomato with a relative population density which were more than 104 and 103 cfu · g-1 respectivelybased on the morphologicalphysiological characteristics and 16S rDNA sequence analysisthe strain HL5-4 was identified Pseudomonas chlororaphis.
