Somatic Embryogenesis from Leaves of Begonia ×Elatior

摘要: 利用丽格海棠盆栽植株叶片培养, 成功地诱导出胚状体并获得再生植株。具体步骤如下: Ⅰ. 在MS + BA 1 mg/L + NAA 4 mg/L + 2 ,42D 1 mg/L 培养基上预诱导10 d ; Ⅱ. 在MS + BA 1 mg/L 培养基上胚性细胞发生胚状体; Ⅲ. 在MS + IBA 0. 5 mg/L 培养基上小植株生根。组织切片观察表明, 胚状体起源于叶片上表皮细胞。

Abstract: Somatic embryo formation was induced from leaves of Begonia ×Elatior by a three-step-method : 1.Embryonic cells were preliminary induced on MS + BA 1 mg/L + NAA 4 mg/L + 2 ,4-D 1 mg/L medium 10 days ; 2.Somatic embryo formation was observed on MS + BA 1 mg/L medium; 3. Roots were induced on MS + IBA 0. 5 mg/L medium. Histological study showed that the embryoid come from upper epidermal cell of the leaf .
