Cloning of Flavonoid-3', 5'-hydroxylase Gene and Its Transformation into Lily ( Lilium longiforum )

摘要: 利用RT-PCR技术从矮牵牛的紫色花瓣中克隆了两个控制花色的类黄酮3',5'羟基化酶基因Hf 1和Hf 2, 并构建了植物表达载体, 利用农杆菌介导法对铁炮百合进行了遗传转化, 获得了转基因植株。

Abstract: Flavonoid-3',5'-hydroxylase ( F3',5'H) is a key enzyme in the biosynthesis of anthocyanins, which can divert the production of delphinidin pigments. Two cDNAs designated as Hf 1 and Hf 2 encoding F3',5'H were isolated from the purp le flower of Petunia hybrida by using RT-PCR and also sequenced. A construct of Hf 2 in a sense-orientation driven by CaMV 35S promoter was constructed and transformed into Lily (Lilium longiforum ) by using Agrobacterium tumefacien mediated method. The transgenic plants were
confirmed that Hf2 gene was integrated into lily genome by PCR and Southern blot.
