  • Regulation of Anthocyanin Accumulation in Grapes in Vitro by Genistein and cGMP

  • It was found in previous paper that the anthocyanin accumulation in peach fruit skin was promoted by genistein(GNT)but inhibited by cGMP,which is quite different from the prevailing hypotheses in this field. In this study,we once again observed that GNT promoted anthocyanin accumulation in grape berry skin in vitro. The anthocyanin accumulation induced by GNT was dependent on light exposure. However,a dark period about 10–12 h was necessary during GNT induction. This may suggest that light is unnecessary for the direct physiological responses for berries to GNT treatment,but necessary for anthocyanin accumulation. Furthermore,it was found that if the berries were treated with cGMP at once following GNT pretreatment,the GNT-induced anthocyanin accumulation was significantly inhibited. However,if cGMP treatment was conducted 12 h after GNT pretreatment,the inhibition of cGMP was no longer observed,suggesting that the GNT-induced physiological responses might be completed in 12 h. Detection by semi-quantitative RT-PCR showed the expression of the genes including PAL,CHS and UFGT,which are related with anthocyanin biosynthesis,were up-regulated by light exposure and GNT treatment in grape skin,while LDOX seemed no response to these two factors. Treatment with cGMP inhibited RNA accumulation of PAL and CHS induced by GNT,suggesting that GNT and cGMP be of antagonism in regulation of anthocyanin accumulation in grape skins.

  • Variation analysis for the quantity and germination characteristics of pollens was carried out in vitro on 92 peach genotypes. Significant variations on the quantity of the total pollens and germinated pollens , germination rate and length of the pollen tubes were observed among the different genotypes, which were estimated to be 0–3 500 grains per anther, 0–2 696 grains per anther, 0–89.33% and 0–1.56 mm respectively, and the model of these data did not correspond with Gaussian distribution. Further investigation on the pollination value of different peaches was performed based on cluster analysis on above data. The population of the cultivars with low germinated pollen quantity and shorter pollen tube length is relatively larger, accounting 45.85% of the total tested cultivars. So the pollination cultivars should be arranged for such cultivars above.

  • Tree growth,yield and fruit quality of eight years old‘Hamlin’sweet orange (Citrus sinensis L. Osbeck)on 7 rootstocks were evaluated in south China. The results showed that the trees with the largest size presented on Carrizo Citrange [C. sinensis(L.)Osbeck × Poncirus trifoliata(L.)Raf.,CT],while the smallest plant crown were on Smooth Flat Seville [C. sinensis(L.)Osbeck × C. paradisi Macfayden,GS],Goutouchen Sour Orange [C. aurantium(L.),GT] and Rich Trifoliate 16-6(P.
    trifoliate,LC). The trees on GS had the highest yield per crown size. The largest fruit in weight and size were gotten from trees on GS. However,fruit from the plants on CT and LC and rootstocks were the largest in number per tree,and fruit from trees on Rangpur Lime(C. limonia Osbeck,RL)had the largest juice content. The largest content of total acids in juice were gotten from the fruit on LC rootstock,the largest content of vitamin C from that on LC and RL rootstocks,and the largest of juice Brix content from that on LC and CT. The largest in T/A(Brix︰acid ratio)fruits from trees on Sunchusha Tangerine(C. reticulata Sunchusa,SM),GT,GS and CT. Whilst,fruit from trees on GT had the largest degree of oleocellosis. Overall,rootstock affected fruit size and weight,juice content,Brix,total acids and Brix︰acid ratio,results so for showed that CT and LC were the most promising rootstocks for Hamlin sweet orange in south China.

  • The resistance of 279 apple germplasm accessions to apple ring rot was evaluated by inoculation with 5 strains of Botryosphaeria berengeriana de Not. f. sp. piricola. Significant differences in the infection rate, the latent period and the diameter of the spot were detected among accessions. The infection rate, the latent period and the diameter of the spot of the same germplasm differed significantly after inoculations with different pathogen strains. The correlationships between resistant parameters were highly significant in response to most pathogen strains. In the screening test for multiple disease resistance, ‘Zhenbao’,‘Gold Sailam’and ‘Jonathan’ performed as comprehensive high resistant, but‘Red Summer’, ‘James Grieve’,‘Shuahong’,‘Ирландское Персиковое’,‘Chaodeng Qiangwei’and ‘Jizaohong’were high susceptible.

  • Youhou sweet persimmon were chosen to investigate the effect of three consistencies of 1-MCP(0.25, 0.50, 1.00 μL · L-1)on postharwest physiology and chilling injury at 4 ℃. The results indicated that 1-MCP treatment efficiently retarded the development of the main chilling injury symptom of sweet persimmon fruit, reduced membrane permeability and MDA content, inhibited ethylene release, but had no effect on respiration rate of the fruits. Further study showed that 1-MCP treatment maintained the higher level of superoxide dismutase(SOD)and catalase(CAT)activities, decreased polyphenol oxidase (PPO)activity and inhibited the increase of peroxidase(POD)activities of fruits. Application of 1.00 μL · L-1 1-MCP treatment had a better effect.

  • High effective simple sequence repeats(SSR)marker based on expressed sequence tags (EST)and the method of bulked segregant analysis(BSA)on the automated fluorescent-labeled system(ABI3130)were employed to screen the molecular marker linked to dwarf gene in kiwifruit (Actinidia spp.) from the F1 separating progenies obtained from the cross between dwarf cultivar ‘Ganmi 5’(♀)and normal cultivar‘Fengxiong 2’(♂)of Actinidia chinensis Planch. Eighty-five pairs of fluorescent primers were designed and screened in the present study. The results showed that a polymorphic band of 285 bp amplified by primer EST-Ad042 was occured both in the dwarf parent and in the dwarf cultivars of separating DNA pools but not in normal cultivar. Furthermore,the polymorphic band had been approved by stochastic amplification in the F1 separating progenies including dwarf and normal genotypes,respectively. The genetic distance between the EST-SSR marker and dwarf gene could be 8.8 cM by genetic linkage analysis. The specific molecular marker of kiwifruit could be effectively mined by fluorescent EST-SSR primers,and the fluorescent primer EST-Ad042 could be utilized to identify its dwarf plant. The screening of molecular markers linked to dwarf gene in Actinidia chinensis Planch had been provided a basis for the studies of dwarf gene mapping and cloning in kiwifruit breeding program.

  • The use of vinegar residue substrate as component of mixed substrate for pepper seedling growth instead of peat was studied. Vinegar residue substrate,peat and vermiculite were mixed in various ratios. Prior to sowing,some physical and chemical properties of the seedling substrates were determined and the growth of pepper seedling was also studied. In most of the cases,the addition of vinegar residue substrate to the seedling substrate produced an increase in total porosity and aeration porosity,a decrease in water holding porosity,whereas the content of vermiculite in mixed substrates increasing,the water holding porosity was also increased;The nutrient concentrations and electrical conductivity(EC)of vinegar residue substrate were higher than peat and vermiculite,less than 75% vinegar residue substrate using in mixed substrate,the plant height, total leaf area and biomass of pepper seedlings were better than control;The addition of vermiculite to the seedling substrate,which had good water holding capacity,the growth of pepper seedling was improved.

  • Cucumber(Cucumis sativus)inbred line NC-46 contains genes that confer resistance to pathogens including Meloidogyne javanica and Meloidogyne arenaria. Hairy root of NC-46 was induced from cotyledon by wild-type Agrobacterium rhizogenes K599 harboring pBIN-35S-GFP with a frequency of 76.7%. A cDNA library of the hairy root of the transgenic cucumber was constructed successfully. The percentage of recombinant clones in the cDNA library was 98.3%. The capacity of the cDNA library reached 2.02 × 106 cfu,and the percentages of full-length gene and Unigene were 47.6% and 66.7%,respectively. Used of the hairy root of the transgenic cucumber for cDNA library construction overcame the difficulty to obtain a large amount of homogeneous natural roots,the varied physiological states of the roots caused by soil microbes,nematode and RNA contamination. With the modified Smart cDNA construction method,the cloned genes in the cDNA library contain the full length of 5′-introns. Moreover,the cDNAs were inserted into a developed pUC19 plasmid instead of λTripIEx2 phage vector. The cDNA containing plasmid can be used directly for hybridization and sequencing,facilitating gene screening. The cDNA library is useful for the screening of pathogen resistant genes and the cloning of genes involved in hairy root development.

  • The taxonomic status, growth promoting and biocontrol effects against Fusarium oxysporum f. sp. cucumerinum of antagonistic bacterium DS-1 strain, screened by co-culture on PDA plates from rhizospheric soil of continuously cropped cucumber in solar greenhouse, morphology, physiological and biochemical characteristics, and 16S rDNA sequencing of the strain were determined, the effects on cucumber seed germination and seedling fusarium wilt biocontrol were studied. The results showed that: The 16S rDNA sequence of DS-1 strain shared 100% homology with Brevibacillus brevis from GenBank. Based on the results of 16S rDNA sequencing, morphological and cultural observation as well as physiological and biochemical determination, DS-1 strain was identified as Brevibacillus brevis, with GenBank accession number EF108303. DS-1 strain promoted germination of cucumber seeds, with significant biocontrol effects against fusarium wilt of cucumber seedlings by 84.92%.

  • Effects of different sulfur level on photosynthetic characteristics,fresh weight and qualities of garlic were studied by nutrient solution. The results showed when sulfur content was 2.25 mmol · L-1,the photosynthetic parameter(Pn,Gs,E),photosynthetic pigment contents in garlic leaf and the fresh weight of bulb were higher. Quality indicators of bulb (except the soluble protein) were increased gradually with plant growth. Among the different sulfur level,the allicin,soluble sugar and free amino acid contents in bulb of garlic and the allicin,soluble sugar,vitamin C and free amino acid contents in bolt were increased by 48.82%,127.92%,63.58%,10.61%,63.30%,29.07% and 74.16%,respectively. Higher content of sulfur(3.75 mmol · L-1)increased the contents of soluble protein in bulb. At a certain range of sulfur concentrations,with the increase in sulfur level,the garlic branching index decreased and then had no significant change with the increase of sulfur level.

  • Combining Ability Analysis on Ornamental Characters of Chrysanthemum

  • The combining ability and genetic parameters were estimated for 7 ornamental characters in F1 progenies of 12 cross combinations of chrysanthemum(Dendranthema × grandiflorum)hybridized by using a 3 × 4 incomplete diallel cross design(NC II)with three cut-chrysanthemum cultivars as female parents and four potted chrysanthemum cultivars as male parents. The results showed that:(1)Flower diameter,ray floret number and ray floret length were controlled mainly by additive gene action,while plant height and leaf length were controlled principally by non-additive gene action. In addition,leaf width and ray floret width might be controlled both by additive and non-additive gene action.(2)‘Zaoyihong’ had a positive general combining ability(GCA)effect for the 7 ornamental characters involved herein,and‘Hereby’could be used as an important parent in hybridization breeding program aiming for preferable double flowers. The three cross combinations,i.e.‘Hereby’בYumantang’,‘Qingxinhong’בZaoyihong’and‘Hongqueshe’ב03-11-2’,performed quite well special combining ability(SCA)regarding mostof the 7 ornamental characters.(3)Variance of population combining ability and its contribution to hybrid progenies suggested that there was a maternal heredity effect for all the ornamental characters studied except ray floret width. The heritability in broad sense(h2B)of the 7 characters ranged from 59.56%(ray floret width)to 80.40%(ray floret number). The heritability in narrow sense(h2N)were lower than 50% except ray floret number amounting to 80.40%. These indicating that ray floret number belonged to high heritability characters and selection pressure should be increased for this character in earlier selection.

  • Effects of GSH on Tolerance to Water Deficit Stress in Cut Rose

  • To investigate the effects of GSH on tolerance to water deficit stress(WDS)in cut rose ‘Samantha’ and the response of AsA-GSH cycle to WDS in petals of cut roses,we pretreated the stem bases of cut rose flowers with exogenous GSH and BSO,the specific inhibitor of GSH key biosynthetic enzyme γ-ECS,to increase or decrease GSH content of cut rose petals respectively. The results showed that GSH pretreatment increased water recovery rate after 24 h WDS and prolonged vase life significantly;On the contrary,BSO pretreatment decreased the GSH content of petals and then,reduced the tolerance to WDS in cut roses. GSH pretreatment increased the total GSH content and reduced GSH content of petals substantially during WDS and water recovery,as well as AsA content. GSH pretreatment promoted the activities of APX and GR,which were two key enzymes in AsA-GSH cycle,but MDA content decreased. Contrary results were obtained in BSO pretreatment. These results suggested that GSH enhanced the tolerance to WDS through the promotion of antioxidant capability of AsA-GSH cycle in cut roses.

  • The flower of Torenia fournieri(Scrophulariaceae)is perfect,usually zygomorphic. Corolla is sympetalous,trumpet shaped and with a lower lip divided into three lobes and a larger upper lip with two lobes. Initiation of five sepal primordia is not consistent in order;The order of initiation is from one adaxial primordium,two abaxial primordia and finally two lateral primordia, while their development is synchronous. Four stamen primordia initiate nearly at the same time before the initiation of the petal primordia and grow slower than the petal primordia. Five petal primordia initiate simultaneously,and two abaxial primordia are fused later. Compared to the floral organogenesis of Agalinis(Scrophulariaceae),there are more differences in the time and order of every floral primordia initiation. While compared to the patterns of floral organogenesis of Rehmannia(Scrophulariaceae),Whytockia,Rhynchoglossum and Titanotrichum(Gesneriaceae),there are not two distinct patterns of the five genera exist in the floral organogenesis,which shows relatively complex relationship between Scrophulariaceae and Gesneriaceae and that the ovary locules and placentation were used to division of these two falimies should be studied further.

  • Analysis of Fruit Flavor Components in Wild Pentaploid Strawberry Native to China

  • Components related to aroma, sugar, acid were assayed in mature fruits of two wild pentaploid strawberry accessions native to China, Hei 1 and Hei 6, by HS-SPME, GC–MS and HPLC. The results showed that the accessions contained 92 aromatic components divided into 11 categories, which included 22 components(5 categories)with relative contents higher than 1%. The predominant aromatic components were acetic acid ethyl ester, butanoic acid ethyl ester, butanoic acid 3-methyl ethyl ester, hexanoic acid ethyl ester, myrtenyl acetate,2-pentanone, 2-undecanone, ethanol,2-heptanol, 1-hexanol, (E)-2-hexen-1-ol, hexanal,(E)-2-hexenal, 2,5-dimethyl-4-methoxy-3(2H)-furanone. Butanoic acid 3-methyl ethyl ester was the unique component in the two wild pentaploid accessions in comparison with Hei 10 (Fragaria orientalis,4x) and Ji 12 (F. mandschurica,2x), and 2, 5-dimethyl-4-methoxy-3(2H)-furanone, 2-pentanone, 2-heptanol, 1-hexanol, and (E)-2-hexen-1-ol were more abundant in the pentaploids. Fructose was the major sugar, followed by glucose, while the least was sucrose. Remarkable difference in sorbitol content was observed between the two accessions. Malic acid predominated in the two wild pentaploid accessions, more than 86% of the total acids.

  • Components and contents of sugar and organic acid in highbush and half-highbush blueberry fruits were analyzed by capillary electrohoresis. The results indicated that average content of fructose was 31.72 mg · g-1,which ranged from 21.38 mg · g-1 to 45.13 mg · g-1 and that of glucose was 31.20 mg · g-1,which ranged from 20.53 mg · g-1 to 45.79 mg · g-1. The ratio of fructose and glucose was
    1︰1.02. Average content of malic acid was 0.61 mg · g-1,which ranged from 0.16 to 3.73 mg · g-1 and that of citric acid 7.31 mg · g-1,which ranged from 1.50 mg · g-1 o 13.13 mg · g-1. Major organic acid in blueberry fruits was citric acid(92.30% of total acid). The coefficient of variation of contents of sugars and organic acids between the blueberry cultivars ranged from 14.25% to 95.45%. Component variation between highbush blueberry was larger than that between half-highbush blueberry. Compared all the sugar and organic acid components,the coefficient of variation of fructose was the smallest,and variation of malic acid was the largest.

  • The perfect coincidence of orfA501-marker and the phenotype of onion cytoplasmic male sterile has arose interesting in researchers. In this experiment,flanking sequences of orfA501 in mitochondrial genome of onion T-type cytoplasmic male sterility was obtained successfully by anchored PCR,and expression pattern of the chimeric gene was analysed by real time PCR. The result showed that:It was cytochrome c oxidase subunit I(coxⅠ)gene who flanked 5′ of orfA501,and a complicated sequence composed by fractions of atp9,atp6,atp1 and nad4 located in the 3′ region of orfA501. Though there were 93 bases between orfA501 and coxⅠ,they transcripted seperately in the anthors of T-type cytoplasmic male sterility,and orfA501 seemed to play an up-regulator factor for the transcription of coxⅠ.

  • In order to study the cross-compatibility between Lagerstroemia and Lawsonia,Lagerstroemia indica cultivars‘Bicolor’,‘XTSC3’and Lagerstroemia caudata were used to cross with Lawsonia inermis respectively. Then pollen shape and stigma form of both parents,as well as pollen tube behavior after pollination were observed. Results indicated that the pre-fertilization barriers caused the sterility of the distant crosses between Lagerstroemia and Lawsonia. Although pollens of Lawsonia inermis were able to germinate on the stigma of any female parents that were used,pollen tube elongating was hindered,which lead to the failure of fertilization in 72 to 96 h after pollination. The incompatibility,when Lawsonia inermis acted as the female parent,was due to the mismatch of pollen shape and stigma form. Moreover,only a small amount of pollens were adhered on the stigma and the germinations were impeded by callose depositon.

  • The species and the distribution of wild Hedychium ornamental resources in China were systematically investigated by the wild work and extensive review of literature and specimens. 26 species,2 varieties,1 form of wild Hedychium were introduced into Guangzhou in open ground and green house separately. The results showed that:There are 32 species,2 varieties,1 form of wild Hedychium plants in China including 2 new species,1 new form and 2 new-recorded species;Some species have specific breeding potential in view of the color of flowers,the type of fragrance and the height of plant in this genus;10 species have wide distribution volumes,but some species with high ornamental values are endangered;17 taxa were successfully introduced into Guangzhou;High temperature in summer is the most important factor to restrict the introduction of Hedychium to Guangzhou. Additionally,some reasonable advices are given on how to exploit the ornamental resources of wild Hedychium plants in China.

  • MADS-box genes encode transcriptional regulators that are critical for flowering and floral organogenesis. To find genes involved in the floral organogenesis of cherry blossom(Prunus lannesiana),cDNAs for one MADS- box genes,ClAP1,was cloned using homologous cloning and 3′ -RACE method based on the sequence database of NCBI. The full length of ClAP1 cDNA is 1 005 bp,containing an open reading frame(ORF)of 717 bp and coding for a polypeptide of 238 amino acid residues. Subsequently, sequence comparison and phylogenetic analysis revealed that ClAP1 was highly homologous to protein AP1 in Arabidopsis and belonged to the euAP1 clade of class A. However,semiquantitative RT-PCR analysis showed that ClAP1 was expressed in sepal, petal, stamen and carpel of three cherry blossom cultivars‘Makino’,‘Hisakura’and‘Albo-rosea’,which was slightly different from the expression pattern of AP1.

  • An efficient plant regeneration protocol for Tagetes erecta L. was achieved via organogenesis from leaf explants in vitro. In this paper,the effects of different combinations of plant growth regulators,the addition of AgNO3,gel and the subculture medium on callus induction and buds regeneration were investigated. The research results indicated that the optimal media for buds regeneration is MS + 6-BA 3.0 mg · L-1 + IAA 3.0 mg · L-1 + sucrose 30.0 g · L-1 + phytagel 6.0 g · L-1 with the buds induction frequency of 97.5%. Put the 13-day-old explants with buds to the tissue culture flask,and 56.7% of them can grow up to 5 cm high after 30 days culture. The rooting of rootless shoots could be accomplished directly in the MS media with rooting frequency of 100%. All of the rooted shoots survived in greenhouse.

  • A two-dimensional electrophoresis(2-DE)system for proteomic analysis of tomato cotyledons was established by optimizing the parameters including the pH gradient of IPG strip,sample application,sample loading and isoelectric focusing conditions. The optimized system include the following steps:Extracting the total protein from tomato cotyledons by TCA/acetone precipitation,separating the proteins with 24 cm pH3-7NL IPG strips,loading protein samples of 300 μg by rehydration method followed by isoelectric focusing programⅠorⅡ,and staining the gels by silver staining after SDS-PAGE electrophoresis. When preparative gel was applied,in order to obtain abundant protein spots,1.5 mg sample was loaded followed by the isoelectric focusing program Ⅲ,and staining with colloidal Coomassie Brilliant Blue. Reproducible profiles with high resolution were obtained by this optimized 2-DE system of tomato cotyledon.

  • ‘Qiumihong’is a new later-maturing peach cultivar which is derived from the cross ‘Okubao’בQiuhuang’in 1980. The fruits are big in size and gorgeous in color. The average single fruit weight is 336 g. The fruits have higher concentration of vitamin C and excellent flavor. The fruits matured in early or middle of September which is the time of Mid-autumn Festival and National Day exactly. Moreover, the flavor of fruits which were stored at 5 ℃ in 20 ~ 30 d was not changed. Therefore the cultivar has a good market prospect.

  • ‘Hongmi Pantao’is a new peach cultivar selected from the flat peach seedlings of‘Zaolu Pantao’grown in solar greenhouses. The fruit development period is about 70 days. The average fruit weight is 144 g,with the maximum reaches to 198 g,and the soluble solids content is 13.2%. This early-ripening peach is flat,white-fleshed,melting sweet,clingstone,and 80% of the skin color is bright red.

  • The new early ripening chestnut cultivar‘Dongyue Zaofeng’belongs to variant dominant strain, which was selected from chestnut seedlings. It has excellent characteristics such as prematurity, good quality, high yield, early fruit, drought tolerance, resistance to barren and pest especially for spider mite. Size of nuts is middle, with flesh fine and smooth. The nuts is perfect for stir-fry, and the new cultivar is appropriate to be planted in mountains and hills in Tai-yi region, and at river beach area in the Southeast of Shandong Province.

  • ‘Jingou 1’is a new early-ripening Chinese dwarf cherry cultivar which is derived from seedlings in 1999,with the average single fruit weight 6.2 g,juice production 82.4%,the content of soluble solids 15.4%,soluble sugars 7.85%,total acids 1.12%,sugars and acids ratio 7.01,calcium 249 mg · kg-1,vitamin C 380 mg · kg-1,and total amino acids 5.13 g · kg-1. The fruits ripen in mid-July in Beijing areas.

The ripe fruit is dark red. It is resistant to phytophthora blight and anthracnose. The fruit is linetype. Fruit length is 25.4 cm. Fruit diameter is 1.69 cm. The average fruit weight is 16.8 g. Fruit flesh thickness is 0.17 cm. The yield of dry hot pepper is up to 4.5 t · hm-2.

  • ‘Qilin’is a new cultivar of Renanthera Tom Thumb propagated by tissue culture of a mature plant selected from the crossing Renanthera imschootiana with Renanthera monachica. The new cultivar is characterized by upright growing, straight plant type, dark green leathery leaves, thick flower stalk with many flowers, long flowering period and long flower life, and strong adaptability. Three years observation on the field trials confirmed the stability of the cultivar. All the above mentioned characteristics indicated‘Qilin’has high ornamental value and good market prospects.
