  • Molecular Cloning and Functional Characterization of MdCBL3 in Apple

  • A gene named MdCBL3(MDP0000155124)was cloned from‘Gala’apple(Malus × domestica Borkh.). The length of MdCBL3 was 852 bp. A phylogenetic tree analysis indicated that the Pyrus bretschneideri exhibited the highest sequence similarity to MdCBL3. The promoter’s cis-acting elements were analyzed and predicted using PlantCare Databases,analysis showed that the MdCBL3 promoter sequence included drought,low temperature,light,auxin,defense responsiveness etc. Construct expression vector of MdCBL3 and use Agrobacterium mediated method to infect apple callus and Arabidopsis seed. Semi RT-PCR indicated that MdCBL3 was genetically transformed into apple callus and Arabidopsis. Phenotypic analysis found that transgenic callus and Arabidopsis seedling grow better and have stronger resistance under NaCl stress compared with contrast. Demonstrated that overexpression of MdCBL3,heterogenous expression of MdCBL3 remarkably increased the tolerance of transgenic apple callus and Arabidopsis to high salinity. The study found that heterogenous expression of MdCBL3 in Arabidopsis enhenced the tolerance of drought and low temperature.

  • Meiosis and 2n Pollen Formation of Ziziphus jujuba‘Wuhefeng’After Treated with Colchicine

  • The microspore mother cell and correlation between meiosis process and flower bud external morphology of Ziziphus jujuba Mill.‘Wuhefeng’treated with colchicine were studied using squash preparation,and the ratio of 2n pollen was determined. The results showed that the first meiosis of microspore mother cell treated with colchicine was normal. Abnormal cytokinesis was observed at the prophaseⅡ;There were four types of spindle body at the metaphaseⅡ,i.e.,parallel type,vertical type, splay type,fusion type,accounting for 27.31%,19.35%,45.27% and 8.06%,respectively. Dyad,triad and tetrad were observed at tetrad stage,accounting for 6.00%,43.04% and 50.96%,respectively. Splay spindle body and fusion body(abnormal-position)were positively correlated with triad and dyad,respectively. The microspore mother cell analyses revealed that 2n pollen resulted from abnormal cytokinesis during prophaseⅡand spindles of abnormal-position during metaphaseⅡof microsporogenesis. And 96.97% of 2n pollen formation was controlled by abnormal-position spindle,which were genetically equivalent to first division restitution(FDR)type of gamete formation that can translate 80% heterozygosity. The other 2n pollens were genetically equivalent to second division restitution(SDR)type of gamete formation which can translate 40% heterozygosity. Thus 2n pollen produced from‘Wuhefeng’treated with colchicine can provide potential value for polyploidy breedingin Z. jujuba Mill. Microspore meiosis process had a corresponding relationship with flower bud external morphology. When the flower bud diameter reached 1.34 mm,microspore mother cells entered into meiosis. The pollen mother cells entered into tetrad period when the flower bud diameter reached 1.68 mm. When the flower bud diameter was more than 1.92 mm,the meiosis was finished and a large number of pollen grains formed. The optimal period of colchicine treatment for 2n pollen induction was at the end of leptotene,when flower bud diameter ranged from 1.34 mm to 1.42 mm. In this situation,the incidence rate of 2n pollen in‘Wuhefeng’jujube substantially increased up to 17.36% compared to the untreated natural one of only 0.

  • Virus-induced Gene Silencing as a Tool for FaMYB5 Gene Functional Studies in Strawberry

  • In the present study,using strawberry cultivar‘Toyonaka’(Fragaria × ananassa) as plant materials,we silenced the gene encoding FaMYB5 transcription factor,which was imparted in flavonoid metabolism in fruits of strawberry by using Virus-induced gene silencing(VIGS)technology. After silencing of FaMYB5,the expression level of other important genes involved in flavonoid metabolic pathway was detected by semi-quantatitive PCR and RT-qPCR. Meanwhile,in order to fully investigate the role of FaMYB5 in regulating the flavonoid metabolic pathway,we determined the anthocyanins and proanthocyanidins content using HPLC and DMACA chemical methods respectively. The results showed that,the recombined pTRV2-FaMYB5 vector and the VIGS system were successfully constructed. By determining of the phenotype and detecting the expression of FaMYB5 after its silencing,the efficiency of gene silencing was about 60%. In constract with the negative control group,the expression of FaMYB9FaLARFaDFR,and FaANR increased,leading to an increasing of proanthocyanidins level. On the contrary,half of the expression level was observed for FaANS,accompanied with the anthocyanins decreasing. While the expression of FaMYB10 had no radical change. In addition,the expression of FabHLH3 and bHLH3Δ decreased after silencing of FaMYB5. However,the expression of FaGL3 had an obvious increasing phase. All the results showed that FaMYB5 transcription factor could negatively regulate the proanthocyanidins metabolism pathway.

  • Characterization and Expression Analysis of Banana MaTIFY1 Transcription Factor During Fruit Ripening

  • TIFY transcription factors(TFs)play an essential role in plant growth and development as well as environmental adaptation. However,the involvement of TIFY TFs in fruit ripening remains largely unknown. In this work,a gene encoding TIFY subgroup member,which was designated as MaTIFY1,was isolated from banana fruit. MaTIFY1 contains an Open Reading Frame(ORF)of 681 bp,encoding a peptide of 227 aa,with molecular weight of 24.97 kD and pI of 7.85. Subcellular localization and transactivation activity assays indicated that MaTIFY1 was targeted to nucleus and owned transcriptional activation activity. qRT-PCR analysis revealed that expression of MaTIFY1 was gradually increased along with the progression of fruit ripening,and was induced by propylene. Moreover,the promoter activity of MaTIFY1 was also activated by ethylene treatment,further supporting the role of MaTIFY1 in fruit ripening. In addition,the recombinant MaTIFY1 protein was available,which provided the basis for further investigation of MaTIFY1’s functions.

  • Nitric Oxide is Involved in Ethylene-induced Adventitious Root Development in Cucumber Explants

  • Cucumber(Cucumis sativus L.)was used to understand the effects of ethylene(ETH)and nitric oxide(NO)on rooting of plant explants. The results showed that the effects of ETH on adventitious rooting in cucumber explants were dose-dependent,and 0.5 μmol · L-1 ethrel treatment was the most effective concentration to promote the adventitious rooting. The root number and root length of explants treated with ETH and NO was significantly higher than that of explants treated with ETH or NO alone. But the positive effect of ETH could be reversed by NO scavenger 2-(4-carboxy-2-phenyl)-4,4,5,5- tetramethylimidazoline-1-oxyl-3-oxide(c-PTIO),nitric oxide synthase(NOS)inhibitor N-nitro- L-arginine methyl ester(L-NAME)or nitrate reductase(NR)inhibitor NaN3,indicating that there might exist synergistic action between ETH and NO on mediating cucumber adventitious rooting. ETH treatments enhanced endogenous NO levels in explants. Meanwhile,the exposure of cucumber explants to ETH up-regulated NOS and NR activity and their gene relative expression. Together,these results indicate that NO may play crucial roles in the adventitious root development induced by ETH.

  • Transformation of Chinese Cabbage Microspores by Particle Bombardment

  • Chinese cabbage transient GUS gene expression was achieved when microspores were bombarded with gold particles coated with plasmid DNA. Helium rupture pressures of 7.58 ×106 Pa and the particle size of 0.6 μm did not significantly affect the viability of microspores of Chinese cabbage. Under optimized bombardment conditions for Chinese cabbage microspores(7.58 × 106 Pa rupture pressure,stopper-to-target-cell distance of 9 cm,0.6 μm particle size,500 μg gold particles per bombardment,and five shots),more than 20 GUS-positive microspores per petri dish were observed after 1 day of culture. The highest number of stably transformed embryos was four per dish in our test results. Collection of microspores of 40 buds,more than 20 transformed embryos were observed. The results of this study suggest that particle bombardment of microspores could be an alternative method for genetic transformation of Chinese cabbage,and may help advance the breeding research and the gene functional analysis of Chinese cabbage.

  • The class B genes,which belong to the MADS-box gene family,play critical roles in regulating the development of petals and stamens in flowering plants. To understand the molecular mechanisms of floral development in garlic(Allium sativum),we cloned and characterized the homologous PISTALLATA/GLOBOSA-like(PI/GLO-like)gene AsPI(GenBank accession number KY272748)from floral buds of garlic cultivar‘Acheng Purple Garlic’. AsPI was 939 bp in cDNA length with an open reading frame of 615 bp corresponding to 204 amino acid residues. The deduced amino acid sequence of AsPI contained a well-conserved MADS domain from amino acids 1–57 and a conserved K box from amino acids 76–140. AsPI had the serine residue and the KHExL sequence within the MADS box and the K box,respectively,a PI motif was present at the C-terminal end,all of which are characteristic of the PI subfamily. The results obtained by sequence alignment and domain analysis indicated that it might belong to PI subfamily of B function genes. Its sequence also formed a subclade with other monocot PI-type genes in phylogenetic analysis. Semi-quantitative and real-time RT-PCR were done to test the expression patterns in different tissues and organs of Allium sativum. The results showed that the expression level in floral buds was the highest,whereas scape followed,little transcripts or no expression was detected in vegetative organs,such as leaf,pseudostem,protective leaf,root. This study developed the positive exploration for preliminary understanding at the molecular level on garlic floral development.

  • Expression Analysis of GA and ABA Signal Transduction Genes in Peaonia ostti‘Fengdan’Seeds Under Different Temperature Treatments

  • Hormone content in Peaonia ostti‘Fengdan’seeds from 8 stages after sowing under 25 ℃ and 4 ℃ treatments was analyzed by ultra-performance liquid chromatography/tandem mass spectrometric(UPLC-MS/MS)method. Four signal transduction genes expression(PoGID1aPoGAMYB4PoGAI and PobZIP)were tested by qRT-PCR. The results showed that‘Fengdan’seeds could germinate under 25 ℃ treatment,while it could not germinate within 100 days after sowing(DAS)under 4 ℃ treatment. The GA3 content increased first and then decreased under two treatments,while it was significantly higher in seed at 17 DAS(seed hypocotyls germination stage)under 25 ℃ treatment than that under 4 ℃ treatment. The ABA content decreased continuously under 25 ℃ treatment,while it increased rapidly after a short period decrease under 4 ℃ treatment,and it was significantly higher than 25 ℃ treatment at 7–17 DAS. It was deduced that the high level of GA3 content at 17 DAS under 25 ℃ treatment could promote seed germination and the high level of ABA content under 4 ℃ treatment inhibited seed germination. The expression patterns of PoGID1a and PoGAI under 25 ℃ and 4 ℃ treatments showed an up-to-down trend,while the expression level of PoGID1a and PoGAI(21 d excluded)were higher and lower,respectively under 25 ℃ treatment than that under 4 ℃ treatment. The expression pattern of PoMYB4 showed an increasing trend under 25 ℃ treatment,while it increased slightly at the initial stages and then decreased under 4 ℃ treatment. On the whole,the expression level of PoMYB4 under 25 ℃ treatment was significantly higher than that under 4 ℃ treatment,at 3–100 DAS. The expression pattern of PobZIP was similar to ABA content change,and it was significantly higher under 4 ℃ treatment than that under 25 ℃ treatment. Therefore,it was suggested that seed germination was controlled by GA and ABA. GA promoted seed germination by positively regulating GA signal ,while ABA inhibited seed germination by positively regulating ABA signal transduction PobZIP expression.transduction PoGID1a and PoGAMYB4 and negatively regulated PoGAI

  • Preliminary Study of Mechanism on Diversity of Floral Organ Variants in Anemone rivularis var. flore-minore

  • After several years’ survey,a serious of nature floral organs variants of Anemone rivularis var. flore-minore were determined. Based on varation organ and varation level,variants were divided into 5 types:white,green-white,five-flap,green and extreme variation. The variation mechanism was studied preliminarily through molecular identification,morphological observations,and AP3-3 genes analysis. The results indicate that both ITS2 and rbcL sequences from normal and variants had no difference after comparison,so the normal and variant plants were identified as the same species. Variant flowers had big differences in morphologys,which existed a variation gradient from outside,and proportion and number of floral organs were all unstable except tepals numbers from normal and five-flap flowers,besides,tepals proportion was negative correlation with stamens/carpels,so it indicated that increased tepals in variant flowers were very likely transformed from stamens and carpels. Based on AP3-3 genes from normal and green-white plants,the rest AP3-3 genes were amplified. Sequence alignment of AP3-3 from normal and variant plants showed that regulation region had a insertion and CDS region had 1 base deletion. Both of insertion and deletion might involve in variation.

  • Analysis of Soil Phosphorus Input and Phosphorus Environment Load Risk in Major Apple Production Regions of Shandong Province

  • Based on the data collection through soil fertility survey of apple orchards,farmer interviewing and agricultural statistics database,the characteristics of P input,surplus and P load were studied using nutrient balance method at major apple production regions in Shandong Province. The results showed that the average application rate of P fertilizer(P2O5)was 676.17 kg · hm-2,and the P through inorganic fertilizer application was 477.36 kg · hm-2(70.60% of the total P input). The characteristic of P input was significantly difference between the three different regions. The highest P input rate(776.52 kg · hm-2)was found in Jiaodong apple production region,and the percentage of chemical phosphorus was 69.14%,which was the lowest in the three regions;the lowest P input rate(348.90 kg · hm-2)was found in Southwestern area of Shandong,and the percentage of chemical phosphorus was 82.95%,which was the highest in the three regions. P surplus was investigated in the 97.82% of surveyed orchards,with the average amount of 407.45 kg · hm-2,and the highest P surplus rate was found in Jiaodong apple production region(615.62 kg · hm-2),followed by Central south area of Shandong(342.81 kg · hm-2),and South western area of Shandong(263.92 kg · hm-2)was the lowest. Soil Olsen-P level in 67.34%,40.51%,and 19.13% in the surveyed orchards had been over the risk threshold level of Olsen-P(50 mg · kg-1)in Jiaodong,Central south area of Shandong,and South western area of Shandong apple production region,respectively. Overall,up to 56.69% of the apple orchards exist phosphorus environment risk in Shandong Province,and the soil Olsen-P content in these orchards was 108.34 mg · kg-1,which 2.17 times of the risk threshold level of P.

  • Incidence and Molecular Characteristics of Citrus leaf blotch virus and Citrus tatter leaf virus Infecting Citrus in China

  • Both Citrus leaf blotch virus(CLBV)and Citrus tatter leaf virus(CTLV)can infect citrus species. To understand the infection status and molecular characteristics of the two viruses in citrus plants in China,citrus samples collected from seven provinces were detected by reverse transcription-polymerase chain reaction(RT-PCR). Results showed that the incidences of CLBV and CTLV were 9.6% and 11.0%,respectively. The partial coat protein(cp)gene of 14 CLBV isolates shared 82.1%–100% sequence identities at nucleotide(nt)level and 88.1%–100% sequence identities at amino acid(aa)level,respectively. The partial cp gene of 15 CTLV isolates shared 89.5%–100% nt and 92.5%–100% aa sequence identities,respectively. Phylogenetic analyzes of partial cp gene sequences of both CLBV and CTLV showed that CLBV isolates identified clustered into two groups. The 15 CTLV isolates separated in different branches,and there is no clear relationships between their phylogenetic positions and original hosts.

  • Phenotype Mutation and Ascorbate Characterization in M2 Population Derived from EMS Treatment in‘Ligeer 87-5Tomato

  • Producing mutants by EMS treatment can create novel germplasm for breeding and novel mutants for gene cloning. In this study,seeds of‘Ligeer 87-5tomato were treated with 1% ethyl methane sulfonate(EMS)for 12 h. 240 plants were investigated in terms of agronomic and biological traits. Parts of M3 plants were verified for genetic stability. Numerous valuable mutant phenotypes were observed in M2 progeny,including premature aging,dwarf,round fruit,low ascorbic acid content,spots,green stem,cluster growth. Taken together the plant growth and fruit quality in M2 fruits,we found 90 mutant plants with 50 mutant traits in total. The 13 mutant plants had two or more mutant traits simultaneously. The mutation rate of all M2 is 8.80%(mutant lines/total lines). The mutant number in leaf,stem,floral organ,fruit and plant habit were 27,7,17,56 and 21,respectively. The mutation rates of leaf,stem floral organ,fruit and plant were 2.64%,0.68%,1.66%,5.47% and 2.05%,respectively. A dwarf mutant gene has been identified as the mutation in curl-3 leading to nucleotide conversion of C → T,and amino acid conversion of L→F. Spraying of brassinosteroid could not restore tomato growth,indicating the curl-3 mutation disrupted brassinosteroid signaling process. The ascorbic acid content in leaves and fruits of curl-3 mutant was significantly lower than those in wild-type,with a higher ratio of DHA/AsA ratio. The expression of AO gene in curl-3 mutant leaves significantly increased. AO enzyme activity of curl-3 mutant leaves was significantly enhanced.

  • Two lectin genes(Plectin1 and Plectin2from Pleurotus ostreatus have been cloned based on rapid amplification of cDNA ends(RACE) method in this study. The full-lengths DNA of Plectin1 and Plectin2 are 1 371 bp,1 359 bp,and contain 5 exons and 4 introns,respectively. The open reading frame(ORF)of Plectin1 is 1 134 bp encoding 377 amino acids,whilethat of Plectin2 is 1 122 bp encoding 373 amino acids. Southern blot confirms both Plectin1 and Plectin2 had only one copy in the genome of P. ostreatus. qRT-PCR analysis shows Plectin1 and Plectin2 were highly expressed in mature fruiting body stage and young fruiting body stage respectively. This suggests Plectin1 and Plectin2 may play an important role in P. ostreatus fruiting body development.

  • Seven populations of Paeonia rockii in Qinling and Ziwuling Mountain areas of Shaanxi were selected to analyze the phenotypic diversity based on 20 phenotypic traits from leaves,flowers and fruits by nested ANOVA,correlation analysis and cluster analysis. The results showed:(1)There were highly significant differences in 20 phenotypic traits among populations. Plant height,Leaf length and seed diameter were highly significant differences within populations. The mean phenotypic differentiation coefficient(VST)of the 20 phenotypic traits was 71.07%,which indicated that the phenotypic variation mainly occurred among populations.(2)The average variation coefficient(CV) of phenotype traits ranged from 7.04% in seed average weigh to 21.66% in seed number in a fruit,which showed that there was low variation of phenotypic traits in Paeonia rockii populations. The average variation coefficient of traits on fruits was 14.45%,higher than that of leaves(13.19%)and flowers(11.40%).(3)Correlation analysis showed that reproductive growth was slightly affected by vegetative growth.(4)The correlation analysis between geo-ecological factors and phenotypic traits showed that the latitude and longitude had significant effect on flowers traits,and the average annual rainfall had the maximum influence on fruits.(5)The cluster analysis on the phenotypic traits showed 7 populations were divided into 3 groups associated with environmental factors,which did not strictly classify according to the geography distance.

  • A segregating population of 296 F1 diploid hybrids has developed from a cross between Rosa chinensis‘Old Blush’(OB)as maternal plant and Rosa wichuriana‘Basye’s Thornless’(W)as paternal plant. In this study,twenty-three SSR markers were found polymorphic within two parents among one hundred and forty-six SSR marks. We selected 94 progenies randomly from F1 population,used twenty-three SSR marks to test the authenticity of hybrids and the genetic relationship. Three homozygous dominant markers(Fv512,Fv609 and 305)were screened from the SSR marker sets,each marker is enough to identify the entire hybrids in our population. In total,all of progenies were identified as real interspecific hybrids,it suggested that the hybrid reliability is very high in our F1 population. Twenty markers were used to examine the genotype segregation,chi-square test indicated that nine(45%)markers exhibited segregation distortion with a significance levels at 5% and 1%,it suggested that F1 population have a high segregation distortion rate and the segregation distortion markers should be excluded when genetic linkage map construction. The UPGMA based dendrograms indicated that the genetic variation and diversity of 94 progenies was significant,and these progenies could be classified into 2 groups and 7 subgroups.

  • Plants produce a series of defense systems under abiotic and biotic stresses,where heat stress transcription factors(Hsfs)are involved as important regulators. Because plants are sessile organisms,plants Hsfs show high diversity in the gene family members,protein structures,and regulatory mechanisms,when compared with the animal Hsfs. Hsfs play important roles in multiple stresses and they are involved in different transcriptional regulatory pathways,which eventually result in forming a complex regulatory network. This review summarized the main protein structures,stresses responses and regulations mechanism of plant Hsfs during stresses.   • ‘Longshuai’is a new apple cultivar derived from the hybrid crossing‘Hanfu’בJinhong’which is high quality,high yield and cold resistant. The fruit is oval in shape. The average fruit weight is 119 g. The fruit having a thin yellow ground skin,covered with red,striped red,and the surface is smooth. The flesh is light yellow,tender and crunchy texture,medium-juice,refreshing and fragrant,sour-sweet. The soluble solids content is 13.9%,the soluble sugar content is 12.75%,the titratable acid content is 5.62 g · kg-1 and the vitamin C content is 128.2 mg · kg-1,with a very good eating quality. The cultivar is suitable for the southeastern of Heilongjiang and the most parts of Jilin Province.

  • ‘Zhuguang’,a pure autotetraploid new cultivar of sour jujube(Ziziphus acidojujuba Cheng et Liu),was induced from‘Xingtai 0604with colchicine by using a novel approach for rapid in vivo induction of homogeneous autopolyploid created in Chinese jujubeZ. jujuba Mill.). Its leaves are dark green with uneven margin. The fruits are big and round,with average weight of 7.76 g and high resistance to fruit shrinking and cracking. The flesh is crisp,sour-sweet,with good taste.‘Zhuguang’is characterized by early-fruiting and high yield. It is an excellent mid-ripening(early-mid of September)table cultivar which is suitable to be cultivated in Xianxian,Zanhuang,Xingtang of Hebei Province and other regions with similar ecological conditions.

  • ‘Zhanggan 40is a new male sterile hybrid variety. Its sterile line is M0105-7,maintainer line is 8513-1-2. Its female parent is the filial generation of sterile line M0105-7 × 8513-1-2 and its male parent is F07013-5. The width of leaf of the variety is 56.08 cm,there are 9–11 outer leaves,the waxiness is light,the head weight is about 1.6–2.4 kg,head height and head width is about 17.0 cm and 14.0 cm separately. The yield is about 75 000–85 500 kg · hm-2. Round ball,center pole is 7.0 cm,2–3 mature outsourcing green leaves,root stock height is 3.0 cm. Tastes crisp and sweet,low taste spicy. Resistance to blastomere and bolting,suitable for planting in the open field in Jibei cold area in high altitude during spring and autumn.

  • ‘Fengyou Youqing’is a new asparagus bean cultivar of whitish-green type and mid-early-ripening,which has been systematically bred from a variant of the‘Jiexi Jiangdou 06-12. Its plant,featuring lustrously dark green leaves,has a strong growth and branching potential. The pod is whitish-green and shaped like a long cylinder. It is 46.6 cm long and 0.93 cm wide,and a single pod weighs as much as 26.1 g. Double pods occur at a high rate,which come out full and even. This new variety enjoys tight and crisp taste,high yield and excellent quality. Because it does not age easily,the harvest can last for a long period of time. Further,the plant is highly resistant to Fusarium wilt,with tolerance to heat,cold and drought. It is suitable for planting in Spring and Autumn in the asparagus beans growing region of Guangdong Province.
