  • A Study on Bud Chilling Requirements of Fruit Trees in Greenhouse

  • Utah model was used for estimating bud chilling requirements of 65 fruit tree cultivars from 5 species in greenhouse during 1995 - 1999. Bud (both leaf and flower bud) chilling requirements were significant differences among different kinds of species and cultivars : grape and sweat cherry possessed the highest chilling requirements (respectively 1 010 - 1 840 c. u , 910 - 1 240 c. u) , chiuing reqairement of plum and apricot were lower (800 - 880 c. u , 790 - 910 c. u) , then those of peach cultivars were the lowest (500 - 910 c. u) . Moreover , the same cultivars had significantly different chilling requirements between different years , it indicated that chilling requirements
    not only were affected by genetic factors but also regulated by enviromental variations. No regular relationship was found between chilling requirements and fruit development period , It also noted that flower bud had higher chilling units than leaf bud in almost all the experimental cultivars. In procssing of endodormancy , root might have some effects and in the experiment it affected the chilling requirements : high root temperature (in fact the temperature was the most suitable for root growth and development) reduced the flower bud chilling requirements , but lower and natural root temperature had no effects. Spraying growth regulators of different concentration also affected the chilling requirements : 10 - 50 mg·L-1 6-BA reduced the chilling requirements and sprouting or blooming , 10 -
    50 mg·L - 1 GA3 increased the chilling requirements , but ABA had no ffects , compared with the control of spraying of H2O.

  • This study aimed at clarifying the effect of pre􀀁harvest fruit cluster bagging on post harvest fruit storability. Adhesive􀀁bonded fabric bags were used to cover Feizixiao litchi fruit. The effects were observed under room temperature, without any chemical treatment and paper box packa ging. Higher water content and lower pH value in the pericarp were observed in bagged fruit with reduced fruit decay rate. Fruit bagging was shown to increase protein content and decrease peroxidase ( POD) act ivity in the pericarp. Thicker waxy layer was seen on the exocarp of bagged fruit and larger cells appeared in the spongy tissue of mesocarp. These changes seemed to prevent the pericarp from losing water and subsequent decay, to delay browning process and thus prolong the storage life.

  • A pair of degenerate oligonucleotides to highly conserved regions at the 3’terminus of L EAFY ( L FY) gene and its homologues , the flower meristem identity genes , is used to amplify a fragment of 883 bp by PCR from the citrus genomic DNA. Sequencing indicated that there is an intron of 473 bp in the fragment . The splice donor site and the splice acceptor site are in identical position to the L FY gene and its homologues. The amino acid sequence deduced from the exons is 77 % - 97 % homologous to that of the corresponding regions of L FY and its orthologues. The expression patterns of
    the citrus L FY homolog ( csL FY) were analyzed by RT2PCR. The result indicates that csL FY expression was detected in vegetative and flower buds from the adult tree as well as vegetative buds from the several month old seedlings. However , the level of the csL FY transcripts in the juvenile buds
    is lower than in the flower buds.

  • A procedure had been studied on cryopreservation of citrus shoot2tips in vitro by vitrification. Shoot-tips of citrus about 10 mm were precultured for 3 days in MT medium supplemented with 5 % DMSO. The excised shoot2tips , 2 - 2. 5 mm in length , were loaded with 60 % PVS2 for 20 - 30 min at room temperature , and were exposed to PVS2 at 0 ℃for 50 - 60 min. Followed by changing the solution with fresh PVS2 , the shoot2tips were immersed into LN directly , and kept for 24 h. After rapid thawing in a water bath at 40 ℃, the shoot2tips were washed with 1. 2 mol/ L sucrose solution for twice and transferred onto MT medium supplemented with BA 1. 0 mg/ L. The cultures were kept in dark for one week prior to exposure to the light . Survival rate of shoot2tips of Poncirus trifoliata was 100 % by TTC examination , and regeneration rate reached to 90 %. The plantlets rooted normally and survived after transplantion. No morphological difference between the regenerated
    plantlets and the control was observed.

  • The effects of soil moisture on the plant growth , yields , physical and chemical properties of strawberry fruit were studied by use of a new auto-irrigation systemof negative pressure. It was showed that plant height , number of leaves , fruit weight , fruit diameter , fruit number and yields gradually decreased with the decreasing of soil moisture. The physical and chemical properties were apparently different under different soil moisture treatments. The Deficit Irrigation decreased water content of fruit , increased fruit hardness , soluble solid content , titratable acidity , sugar-acid
    ratio and the content of ascorbic acid. The correlation analysis indicated that soil moisture has a positive correlation with the fruit diameter and weight , but has a negative correlation with soluble solid content and sugar-acid ratio.

  • The content of DNA, RNA and total nucleic acid ( TNA) in stem apex increased
    gradually before the inflorescence differentiation, and reached a peak at the inflorescence differentiation, then decreased to a proper level at one week after the inflorescence differentiation and increased again to form little peak at the former stage of curd formation, then decreased again at the late stage of curd formation. The RNA/ DNA rat io went down rapidly just before the emergence of curd, and went up gradually from the emergence to harvest of curd. The content of Calmodulin ( CaM) in stem apex went up gradually before the inflorescence differentiation and reached a peak after one week of the in florescence differentiation, then went down during curd format ion. N-Zn nutrition could promote the metabolism of nucleic acid and CaM, but different N-Zn nutrition had different effects on the synthetic activity of nucleic acid. Suitable N and Zn nutrit ion had a marked promoting effect on the synthesis of CaM.

  • The genetic effects of node of 1st female flower , fruit number per plant , individual fruit weight , fruit length , fruit width , pulp thickness in Momordica charantia L. were analysed on the data from the parents , F1, F2, B1 and B2 of two cross as ( ZH967281 ×G968031 and ZH961123 ×G968031) by using three inbred lines with significant difference. The results showed that genetic patterns of six economic characters all accorded with the pattern of additive-dominance and additive effect was major. Node of 1st female flower , fruit number per plant , individual fruit weight , fruit length and fruit width were all positive dominance partially and tended to the larger parent , but
    pulp thickness was negative hyperdominance and tended to the smaller parent . The narrow heritability of node of 1st female flower , fruit number per plant , individuam fruit weight , fruit length , fruit width and pulp thickness were 69. 23 % , 61. 94 % , 61. 64 % , 76. 18 % , 28. 98 % , 35. 09 % ,
    respectively.

  • Effects of several factors such as medium , cutting materials and plant growth regulators on in vitro culture and rapid propagation of Opuntia milpa alta Haw. were investigated. Using the Opuntia milpa alta Haw. stem as explant , a method for high frequency propagation of Opuntia milpa alta Haw. was established. The results of the experiments suggested that the appropriate medium for different culture stages were : (1) Germination of adventitious buds : MS + 6-BA 3. 0 mg/ L + NAA 0. 1 mg/ L. On this medium shoot multiplication occurred at a 82fold rate after 30 days ; (2)
    Differentiation and subculture of rosette buds : MS + 6-BA 1 mg/ L + KT 0. 5 mg/ L + NAA 0. 3 mg/L ; (3) Rootage : 1/ 2 MS + NAA 0. 1 mg/ L + IBA 0. 1 mg/ L. The shoots could be directly rooted in transplantation medium (soil , vermiculite and sand in equal proportion) with up to 95 % survival .

  • Genome relationships of Chinese Cabbage species were studied by molecular marker technology AFLP. 4173 bands were detected with 10 pairs of primers. Clustering analysis showed that it is distant between turnip and Chinese Cabbage ; Jiguancai (Brassica campestris L. ssp. chinensis Makino) and Maobaicai ( Brassica campestris L. ssp. chinensis Makino) are close to head Chinese Cabbage ( B . campestris L. ssp. pekinensis Olsson) which is distant to other non-head Chinese Cabbage varieties. The relationship among non-head Chinese Cabbage (B . campestris L. ssp. chi-nensis Makino ) varieties including Taicai , Caixin , Chaiwu , Jianganbai and Ma’erduobai was
    analyzed on molecular level in this paper , which provides some information of origins and evolution of Chinese Cabbage.

  • 1% fermentation matters from four weeds ( Artemisia princeps Pampan, Cayracia jap onica Gagn, Ste llaria media Villars, Trifolium rep ens L. ) were respectively added into pot soils which contained high density propagules
    of vesicular􀀁arbuscular mycorrhizal fungi for culturing trifoliate orange ( Poncirus trifoliata Raf. ) seedlings. After 70 days, mycorrhizal infection rates were high in all treatments. But compared with the control, fernentation matters signi􀀁ficantly stimulated the growth of trifoliate orange seedlings, reduced the mycorrhizal formation in roots of seedlings, and there were lower infection rates in treatments of fermented weeds than in that of fermented liquids; High concentration of nitrogen also could reduce the mycorrhizal formation level; The phenomena of parasitism and growth depression in mycorrhizal citrus took place when the mycorrhizal infection rate was too high.

  • ‘Redchief’and‘Red Fuji’apple tree ( Malus domestica Mill. ) were used as materials to test for the effect of treatments of paclobutrazol ( PP333 ) and GA3 on the contents of endogenous hormones in buds. The results
    showed that the contents of and ZR , and ZR/ IAA , ZR/ GAS , ABA/ IAA and ABA/ GAS ratios in spur bud were remarkably increased , PP333 treatment by while GAS and IAA content in spur bud were decreased. The contents of GAS and IAA in spur bud were remarkably increased by GAS treatment , while ZR and ABA content , ZR/ IAA , ZR/ GAS , ABA/ IAA , and ABA/ GAS in spur bud were decreased. As to the effects of spraying PP333 or GA3 , it was noted that PP333 could increase the leaf bud node number , but no effect was noted for flower bud ; GA3 had no effect on the node number of leaf bud , but had a trend to decrease the node number for flower bud.

  • Various explants of ‘Yulu’peach including leave , young shoot , hypocotyl , ovule , ovary , endosperm, immature embryo collected approximately 45 - 50 days or 75 days after full bloom , and mature embryo were cultured in vitro. As a result , only the white nodular callus derived from immature embryos collected 45 - 50 days and 75 days after bloom succeeded in producing adventitious buds , and the regeneration rates of white callus were 73. 8 %
    and 15. 5 % respectively. 62BA had stronger effect than KT in inducing adventitious bud formation. Adventitious shoots were transferred and cultured to plantlets. Effect of different hormone , carbon source , medium and transfer time on redifferentiation of adventitious buds from callus derived from immature embryos have been discussed.

  • The rooting technique of fruit tissue culture system with filter paper bridge were studied in this experiment. The results indicated that the rooting rates of Zhumei crabapple and strawberry with filter-paper bridge were 20 %
    and 4. 1 % more than those of their controls respectively ; the rooting rate of cherry stock was same as that of its control , but the rooting time was decreased to 3 - 10 days ; the transplanting survival rates of these three kinds of fruit tree plantlets all increased up to 93 % , the survival rates of Zhumei crabapple , strawberry and cherry stock were increased by 16. 1 % , 21. 4 % and 14. 9 % respectively , compared with their controls ; the root vigor areas of Zhumei crabapple , Strawberry and cherry stock were increased by 6. 9 % , 11. 54 % and 7. 5 % , compared with their controls ; the thickness of epicuticle , hypodermis and pajisade tissue and the leaf stoma density of Zhumei crabapple were increased by 45. 8 % , 50. 3 % , 47. 6 % and 2. 5 % respectively , compared with its control. This method is simple and convenient. The cost of tissue culture could be cut down 75 % than traditional rooting methods.

  • ‘Nuomici’Litchi trees (82year-old) were sprayed with calcium and gibberllin and the control trees were sprayed with water. The results showed that the treatment with Ca (NO3) 2 and treatment with GA all could reduce
    the proportion of cracking fruit distinctly , In the early stage , the content of water2soluble pectin and protopectin increased , meanwhile the content of protopectin increased more than the content of watersoluble pectin , but when the fruit was becoming ripe , the content of watersoluble pectin decreased gradually while the content of protopectin increased largely. PE and PGactivities tended to increase with fruit development till ripening stage when they turned to decrease sharply. There was significant correlation between PG activity and the content of water-soluble pectin. CX increased throughout the whole development and has two peaks.

  • The surfaces of Valencia orange fruits with natural and commercial wax coatings were examined by the scanning electron microscope (SEM) , and the effects of natural wax , washing , wax removal and waxing on weight loss
    of fruit were also discussed. The results show that the natural wax platelets on the surface of fruit are irregular in shape and size. Most natural platelets can be removed by dipping the orange in a chloroform-methanol solution. Removal of natural wax platelets resulted in heavy loss of water in the fruit. Waxing partially or completely plugs stomatal pores and effectively reduces the weight loss of fruit.

  • The flavone contents of Ginkgo biloba leaf stalks and leaves in different types of the branches and leafpositions were assayed in 1999. The results indicated that the flavone content rested on long-branch leaves had three
    peaks , but only two peaks in both middle-short branch leaves and phyllome-branch leaves. Of all the peaks , the last one showed the highest value in all types of the branches. At the same time , the increasing speeds of flavone contents queued from fast to slow: long-branch leaves , middle-short branch leaves , phyllome-branch leaves and the leaf stalks. However there was an opposite trend of the change of flavone content in the leaves and its leaf stalk during the late stage. Flavone content in the middle-position leaves of long2branches was the highest , that in the basal leaves was the second ,
    and that in the top-position leaves was the last. The relationship of the change of flavone content and its distribution with leaf classification , collection and trimming were discussed.

  • This paper reports the studies on in vitro culture of shoot-tip of illered-onion , including callus induction and differentiation , plantlet root development and transplantation. Results showed : (1) MS basal medium supplemented with 2 ,4-D 2. 0 mg·L-1 and KT 0. 5 mg·L-1 was advantageous to callus induction. The frequency of callus induction was 100 %. (2) The combination of NAA 0. 1 mg·L-1 and BA 0. 4 mg·L-1 was the best for callus differentiation with the ratio of 88. 3 %. Most of callus was aggrandized again and induced green bud spots then formed multiplying clumpy buds , the mean number of plantlets per callus was 10. 2 . (3) 100 % of the regenerated plantlets treated with NAA 0. 01 mg·L-1 and IBA 1. 5 mg·L-1 and PP333 0. 1 mg·L-1 could develop their roots , grew robustly and robust green leaves , after the regenerated plantlets were transplanted to soil. The survival ratio of transplanting reached 100 %.

  • Raphnus sativus-antifungal proteins ( Rs-AFPs ) are cysteine􀀁rich, antifungal proteins. The Rs-AFP1(2) genes were inserted into the vector pTrxFus and transferred into E. coli GI724. The fusion protein from recombiant pTrxAFP1( 2) was about 22 kD and taking up 0. 4 and 0. 5 percent of the total proteins respectively. Inhibition experiment showed that the expressed products had antifungal activity to Colletotrichum musae. Expressed products from pTrxAFP2 showed much stronger inhibitory activity than that from pTrxAFP1. A plant expression vector pBIAFP2 was constructed, which had been transferred into tomato by Agrobactrium mediated transformation. 32 plantlets had been obtained and 28 of them detected by PCR and Southern Blot. The results showed that 17 of them were positive, and the results of Southern Blot showed 6 of them were positive.

  • The genomic diversity of thirty-five tree peony cultivars of 7 different flower color were analised by RAPD. The results showed that 337 (80. 62 %) polymorphic sites were detected among the total 418 sites amplified indicating abundant genomic diversity of tree peony cultivars. Similar results were also revealed between accessions within the same flower color series , but significant differences were observed between different flower color series in the order as Blue series > Yellow > Dark red > Black >White > Green > Red ones based upon the percentage of the polymorphic sites.
    The genetic distance and genetic relationships of the accessions were constructed through unweighted paired group method with arithmetic averages cluster analysis , all accessions were grouped into 5 genetic types , no significant correlation between flower color series and genetic types were detected except Red flower series.

  • ‘Yuanling 1’was selected as a high nutritious clone applicable to drying and processing from Yuanling cultivar. The fruit size is big and uniform with the average fruit weight of 16- 18 g and the soluble solids content of
    33%, the dried fruit is high quality and drying rate of fruit is about 60%. It is early bearing, its yield is 30% higher than Yuanling , and fruit drop before harvest is less. The critical low temperature of fruit set is 22- 23at blooming.
