  • Physiological Reaction on Resistance to Iron Chlorosis in Malus xiaojinensis and M .rockii

  • Malus xiaojinemis seedlings grew normally without any iron ehlorosis symptom at soil pH 7.8 and its leaf chlorophyll content was much higher than that of control species M.rockii. which showed severe ehlorosis under the same pH conditions.Mild but typical iron ehlorosis symptom was observed in scion leaves of M.xiaojinemis grafted on M.rockii rootstock in the field with pH 7.8. Conversely,when M .rockii was exploited the scion and grafted on M.xiaojinenesis rootstock, its iron ehlorosis symptom disappeared.The rate of root H+ excretion and ferric-chelate reduetase activity inereased in M.xiaojinemis and were substantially higher than that of M.rockii under iron deficiency.The part of root H+ excretion overlapped with the functioning region of root FCR, It is hypothesized that the high root H+ excretion in the absorptive parts in the roots of M.xiaojinemis provided an optimum pH environment for root FCR.

  • The contents of total sugar, sucrose, ducrose, fructose and the activities of sucrose-metabolizing enzymes containing acid invertase, sucrose synthase, sucrose phosphate synthase and amylase in the fruit flesh of Juglans regia L.were analyzed. The results showed that the highest content of total sugar was found in the late developmental stage, with the starch content being varied during fruit development an d contents of soluble sugars changing according to the kinds of sugars. The fructose content gradually decreased in the late developmental stage, but the contents of sucrose and the ducose increased.The activities of acid invertase and am ylase were higher at the early developmental stage than at the late developmental stage; and the activities of sucrose synthase and sucrose phosphate synthase showed a similar chan ge pattern with lower activities at the early developmental stage and higher activities at the late developmental stage.

  • Loquat trees(Eriobotrya japonica Lindl.)grown in two open-top growth chambers at CO2 concentrations of 400 μmol·mol-1 and 700 μmol·mol-1 were used to study the effects of elevated atmospheric CO2 ,concentration and water stress on photosynthesis.chlorophyll fluorescence and acting of an tioxidative enzymes in leaves.The results showed that elevated atmospheric CO2 concentration(700 μmol·mol-1) significantly enhanced the photosynthesis under various levels of water stress. Furthermore,under high CO2 condition, the values of Fv/Fm , Fv/Fo and PSⅡ were also higher than at normal atmospheric CO2 ,concentration,but the diference between the aterstressed
    plants and well—watered plants was not significant. In addition, elevated atm ospheric CO2 concentration considerably jncreased the ach vities of SOD, POD an d CAT. Such jnerease, however, was farless significan t liBder water stress condition.The MDA content also showed less increase under the latter condition. The results indicate that high CO2 concentration seems to have ameliorating effect on drought—induced oxidative damage.

  • The effects of salt (NaCl) stress on diurnal course of photosyntIlesis of young ‘Hosui’ pear trees on two rootstocks with different salt tolerence。Pyrus betulaefolia and P.caueryana were studied by portable photosynthesis system CIRAS-1 with pot-grown grafting seedlings in soil salt-containing 0.3%,0.6% or no(control).The results showed that under natmal conditions(control),diumal course of net photosynthetic rate(Pn) in pear leaves presented two peaks,and daily variation of stomatal conductance(Gs)and transpiration rate(Tr)applied single peak;while salt stress resulted in changes of diumal variation of photosynthetic properties,especially whole decline of diurnal curve of Pn;and changes of diumal curve of photosynthetic chareactefistics(or the decline of Pn)were sharply afected by diferent rootstocks or salinity as wel1.On the other hand.under salt stress,diurnal variation of Tr had linear relations with variation of Gs in leaves of pear tres with rootstock P.betulaefolia,while diurnal variation of Tr had linear relations with variation of VPD in pear trees with rootstock P.caueryana .In addition。in the forenoon or aftemoon (under rather suitable environment)stomatal limitation was main inhibition factor ofphotosynthesis of‘ pear,while in midday(under higher light intensity and temperature and arid air) nonstomatal limitation was tnain inhibition factor.

  • By adopting complete diallel crossing method, inheritance of clubroot resistance in cabbage (Brassica oleracea L.var.capitata L.)was studied in the seedling. The results showed the resistance was controlled by at least three pairs of nuclear genes, and was imcompletely recessive. Effect of backcross was very significan t and that of reciprocal cross was not. Resistance Was dominated by nuclear gene, and efect of cytoplasm was very small. Narrow heritability was comparatively high.Both general combining ability (G.C.A)and special combining ability (S.C.A)were very significant.The resistance comfonned to the additive-dominance model, and dominance was dominating. To some degree, the resistance to clubfoot disease showed a quan titative
    character.

  • The male sterile line of Chinese cabbage CMS3411-7 and CMS7311, with cytoplasm of Brassica campestris L.were induced under 5 /9 days condition to obtain trace pollens, which relative vigor an d cross-fertile abihty was tested by biological way. Its results show that the quan tity of trace po len in an ther of sterile grade 1,grade 2 and ade 3 were 73.5%-84.4% ,47.6%-51.5% (half sterile)and 28.4%-32.5% of normal anther respectively; the trace pollen in an ther of sterile grade 1 and grade 2 both Can fertile by self-polinated and got about 3.9 grain seeds per silique, which is similar to the situation of low incompatible line; the cross-fertile ability of it is only 9.1%-6.6% compared with po llen of flowering Chinese cab bage (Brassica campestris L. ssp. chinensis L.var.purpurea Bailey) even pollinated 4 h before.This is about one sixth to one tenth of normal pollen.Additional, we present the qu estion that should be paid attention to in hybrid production, and the reliab ility of using the trace pollen induced to reproduce CMS lines was discussed.

  • Abstract: cDNA—AFLP was used to detect diferentially expressed genes during flower bud development of
    male sterile an d fertile lines of cabbage an d broccoli. Expression profiles were generated around 26,000 cDNA frag—
    ments(128 primer combinations).Transcriptional changes were observed for 24 cDNA fragments,from which,
    1 3 were only expressed in fertile buds, while the other 1 1 were upregulated in fertile buds. Sequencing of a 300 bp
    diferential fragment amplified by primer combination A16T15 revealed 89% similarity to the gene thioredoxin,
    which is related to pollen development in the Brassica family.

  • An eficient an d reliable technique Multiplex— CAPS was developed for genetic characterization by
    combination of two principles Multi—PCR and CAPS (Cleaved amplified polymorphic sequence) in tomato.This
    techniqu e included (1)using 2 or 3 or more than 3 primer pairs in the same am plification reaction,after 30—40
    thermal cycles the PCR yield diferent products which correspond to each individual primer pair, (2) using only
    one enz、111e to digest these diferent PCR products, (3)Using agrose gel electrophoresis to detect polymorphisms
    followed by ethidium staining. By comparison with CAPS, Multiplex— CAPS Can detect the polymorphism of 2 or 3
    or more than 3 primer pairs in the same am plification reaction at the same time. Th e polymorphism of Multiplex—
    CAPS equal exactly the polymorphism supe rimpose of each individual primer pair. Multiplex— CAPS has good re—
    producibility with high an alytical efect, lOW cost, lOW labor an d time consuming.

  • Abstract:Single PCR reaction with two SCAR markers respectively tightly linkedwith M/an d Sw一5 genes in tomato,which resistant to root—knot nema tode an d toma to spotted wi lt virus,has been used to screen the multiplex
    ban ds.The PCR products with multiplex primers were completely correspond to the amplified bands produced by single SCAR primer.Among them,codominant SCAR 1 Balker tightly linked with Mi gene produce 750 bp flag—
    ment in both resistant an d susceptible toma to lines.Th e am plified ban ds from susceptible an d resistant lines were distinguishable after cleavage wi th the restriction enzyme I. Genotype wi th homozygous an d heterozygous M /
    gene could produce respectively 570 bp, 160 bp an d 750, 570 bp, 160 bp bands . Susceptible genotypes still present 750 bp fragment.The dominant SCAR2 marker tightly linked with Sw一5 gene would produce only 400 bp PCR product in resistant genotype. Th e replicated stable results pmved that tw o resistant genes could be identified simultaneously by using corresponding SCAR primer under adaptable condition.This system compared with single
    primerPCR would be time saving, less labor an d low cost. It could be very useful for marker—as sisted selection during early stage in tommo and fficiently speed up breed ing procedure.

  • T0tal RNA was extracted with an impmvod one.step method from tobacco leaves infec ted with potato
    virus X (P、Ⅸ ).The mole quality of tested RNA was very good,which indieated that the impmvod method was
    very efective and simple. A pair of specific primers were designed an d synthesized based on the homologous
    nueleotide sequence of diferent isolates of PVX by the reverse transcription an d polymerase chain reaction (RT.
    PCR),a 308 bp DNA fragment was amplified successfully from PVX of Shandong isolate,and no fragment was
    obtained from contro1.With only 1U Reverse Transefiptase XL (AMV),the am plified resnlt were still very good.
    The resnlts showed that an economical an d convenient RT.PCR detec tion system of Pvx was set up.

    He, naturally distributed in the northem subtropical area of Yan gtze River.This study was done in late autumn of
    2002 in Nanjing Botanical Garden,to evaluate the pollination biology of C.nitens Oliv.The pollination charac teristics including flowering phenology, flowering pattem , species of insects, visitation rate of visitors and their
    foraging behavior were investigated,and pollen viability,stigma receptiblity polen—ovule ratio (P/O)and breed—
    ing system were also studied .The results show that C. nitens Oliv.is an an thophilows species;its breeding sys—
    tern is facultative hybridization,as the P/O is about 1 190 according to Cruden (1977).The efect of the wind to
    spreading po len is little.an d beetles&re its main visitors which can cause serf-fertilization,scarcely any be es and

    during vase were improved obviously by three types of treatments compared with control, such as it is increased in

  • The high frequency regeneration system of carnation from leaf to callus to plantlets was established.
    Then genetic transformation of the carnation with the ACO an tisense gene.which controled by flower-specific expression
    promoter(PchsA)was performed via agrobacterium mediated method and obtained 3 of transgenic plants
    via the pathway of regeneration plantlets from callus at last.Th e resuhs of the PCR and PCR.Southem hybridization
    analysis showed that the carnation ACC oxidase antisense gene Was integrated into carnation genomes.It will be a
    good foundation for improvement the longevity of carnation flowers.

  • Abstract:The results of semi vitro culturing pollinated styles excised from plan ts indicated that the amount of
    po llen tubes from style bottoms and the percentage of styles with po len tubes through their bo ttoms in all tested
    styles difered wi th the CroSS corotfinations including the incompatible combination an d compatible comb ination.For
    the former ones, the percentage ran ged from 0 to 32.4% , most of which were under 16% , while for the latter
    ones. the percentage ranged from 51.6% to 86.7% that was ClOSe to the rate of field fruit setting after CroSS-polination.
    Furtherm ore. when the pollen germination rate Was above 20% , there Was no significan t diference between
    the flowers from the room water cultured flowering woods an d field orchards as for the percentage of styles with po len
    tubes through their bottoms in all styles.So we can extend the time span of collecting materials for our experiments.
    For the first time,the method of semi vitro culturing po llinated pear styles including CroSS compatible an d CroSS incompatible
    Was repo rted.We have also identified the genotypes of pear cuhivars Chikusui, Kisui an d Aikansui,
    nam ely 5354, $4S5 an d $4S5, respe ctively.

  • ‘Huanghua’ and ‘XiaJ1 aJ1’ pears that were significan tly diferent in storage an d texture trait
    were used to investigate contents of Ca2
    , total calcium in flesh, seeds an d pe ctin, the results showed that:
    1) Contents of Ca2 and total calcium were the highest in young fruit stage,then they decreased along with fruits
    development.Content of calcium in flesh of‘Huanghua’was significantly higher than that of Xiangnan before
    harvest;2)Total calcium content in seeds Was much higher than that in flesh。and it increased along with fmits
    development gradually.Content of total calcium in seeds of‘Huanghua’kept rising after rapid expanding stage of
    middle July, while that of ‘~ an gnan ’ stopped rising after late July. A Ca2 rapid rising peak in seds of two
    eultivars appeared in late fruit development;3)Content of proto pectin increased first,and then it began to de—
    crease.The period when proto pectin in ‘Huanghua’stopped increasing was earlier than that of‘Xiangnan’.
    and proto pectin of‘Huanghua’Was significantly lower than that of‘~angnan’in late fruit development.Watersoluble
    pectin contents of two eultivars increased gradually,and they were adjaeent.

  • Ginkgo pollen iS scanned and transmitted under electronics microscope an d something iS foun d:
    (1)The ginkgo pollen grain form,for the boat shape under the dry condition,it is back for the oval to inhale water,
    and it is compose of two parts,the sprouting thin-wall construction (occupying about 40% of total area)and
    the ectotheca with overburden(occupying about 60% of total area), and both border land have clear warty evection.
    The form is NoP1C1 type in NPC system on the classification. (2)The warty ornamentation polen has the
    three kinds,Smooth type,rough type and I-type. (3)The uhrastmcture of polen under the electronics microscope
    shows:The ectotheca ofginkgo polen is composed oftwo part,the sprouting thin-wall construction(0.23—
    0.28/an in thickness)and overburden(0.75—0.82 pan in thickness).The overburden in border land of them iseleadv

    thicken to warty eveetion. The lacuna among the baculums is linked together with milipore. Th e sclerench~

    ana of overburden, spreads when inhaling water and uncovers sprouting part, an d systoles when losing water
    and covers sprouting part partly,which preserves the sprouting part. (4)Th e only reproduction cell, famess,
    l0.32一l1.28 lan in length,is 2-cellular type. It is nearby situated the equatorial axis, an d parallel th it in

  • To provide the theoretical basis on walnut cultivation, the water contents an d the photosynthetic
    rate of walnut leaves were studied after the leaves were treated with natural brassinolide. The experiment Was set up
    as a randomized complete design th three replicates.The research results showed that after walnut leaves were
    sprayed with diferent concentration of natural brassinolide, leaf water content, relative water content, free water
    content an d critical saturation deficit were increased. Natural saturation deficit, the degree of water requirement,
    the rate of transpiration,cell membrane penetration and injury rate were decreased,and chlorophyl content and
    the rate of photosyn thesis were raised.The efect of0.30 mg‘L一 on walnut among the diferent concentrations of
    the natural brassinolide Was the most obvious.Compared with control, leaf water content, relative water content,
    free water content and critical saturation deficit were increased by 9.6% . 16.1% ,7.9% and 26.8% respective.
    1v.Th e natural saturation deficit an d the degree of water requirement were reduced by 9.0% an d 24.5% .Th e rate
    of transpiration at 1:30 P.m.was cut down by 32.7% .The cel membrane penetration and injury rate were

  • In this paper, effect on respiration, ethylene production, soluble solids and content of carbon dioxide the fruit core of‘Yali’pear using一100 kV/m high—voltage static electric field (HVEF)were studied.The results showed that HVEF had significan t effect on the respiration with a 60 d delayed peak and ethylene wi th a 60 d delayed pe ak but a half value compared to the control.Compared wi th control fruit,Furthermore, HVEF reduced loss of solub le solids of ‘Yali’ pear and inhibited browning of the fruit. To restrain function of respiration
    caused by HVEF may msulted from a metabolized turbulence of Fe2 /Fe3 an d restrained electric transfer of respiration chain.

  • Two monosomic alien addition lines 02.17 and 02.39 (2n:15. 14C+1H)were identified among 52 plan ts derived from embryo rescue of an Cucumis allotriploid× cucumber. Diferent from their allotriploid parent, the plan ts have white spine, an d the shape of leaves in 02.17 an d 02.39 was has tate an d palmate respectively.In pollen mother cells (PMCs) of 02.1 7 and 02.39.7 bivalents and 1 univalent were observed at diakinesis and metaphase i.Low frequency (about 4% 一5% )of trivalents has also been observed at metaphase JI.At metaphase 11,8 chromosomes went to one pole while 7 to another.R APD analysis indicated these two plants were added with one alien chromosome from C.hystrix.

  • Germination efects on backwheat nutrition was studied. The results showed ‘Yuqiao 1’ (FagoloTum esculentum)and ‘Yu 6—21’ (F.tartar/cure)seeds contmned large an3ount of trypsin inhibitor After ten days germination there Was almost no trypsin inh ibitory activity in buckwheat seedling. Amino acid analysis showed that the essential amino acid of sedling were more equilibrium than that of the seeds and score of ratio coefiient of amino acids(SRC)of‘Yu 6—21’was approximate to egg’S,The SRC of ‘Yuqiao 1’slighfly surpassed that of egg’S.The SRC of seedlings Was higher than that of seeds.The seedling contain high level of vitamins and rutin content increas ed by four to six folds after 10 days germination. All evaluation results showed germination improved the nutrient value of buckwheat.

  • Gas exchan ge an d chlorophyll fluorescence were used to explore the photosynthesis of Areca triandra Roxb. in greenhouse before and after partial shading. It showed that light saturation photon flow density of Areca triandra Roxb. Was about 200—300~tmol·m一 ·s一 and optimal leaf temperature Was about 25℃ .The average daily photos yn thetically active solar irradian ce decreased by about 55% , while for the air an d leaf temperaturea decrease of about 3—4℃ were observed after shading compared with before shading.Distinct photoinh ibition of Areca triandra R0xb. Was observed before shading, however, there Was no obvious photoinhibition occurred after shading. The average daily net photosyn thetic rate of Areca triandra R0xb. Was only reduced by about 12% after shading. All these results indicated that partial shading Was be nefit for the growth of Areca triandra R0xb. in greenh ouse in summer.

  • Oncidium aloha ‘1wan ga’ in vitro shoots were used as plan t materials for experiments to study the efect of difierent sucrose concentrations (30,20,10,5,2.5,0 g·L )in the culture medium on the growth 0f the in vitro sho ts under CO,enrichment conditions. The results showed that the in vitro shoots had photoau totrophic capability. Under CO2 enrichm ent conditions, in vitro shots being cultivated in the sugar-free culturemed ium could grow normally by photoautotrophic capab ility,but a bit slowly. Increas ing the sucrose concentration in the culture medium could significan tly promote the growth of the in vitro shots of Oncidi. Under CO2 enrichment condition. in vitro sho ts grew be tter in the sugar culture medium than in the sugar-free culture medium .Compared th the sucrose concentration (30 g·L )un der normal culture conditions ,when the sucrose concen tration dropped to 20 g·L|。. there was not significan t difference be tween the tw o concentration treatments in the main growth indices of the in vitro shots including plant height, length of leaves, range of leaves, number of roots. total fresh mas s an d dry matter mass. The above results indicate that under C02 enrichm ent cul ture conditions. the existence of sucrose in the culture medium is still necessary for Oncidium ,but compared with normal culture methods. it can partly reduce the amount of sucrose used in the culture medium .

  • The bulbs of tuberose were treated with 6~Co.7 ray in dose of 10 Gy,20 Gv,30 Gv,40 Gv and 50 Gv respectively and cuhivated in andomized blocks design after treatment.1he untreated bulbs used as control.Survival rate and the height of plant were examined.The result indicated that it Was obviously that the inhibition of both the survival rate and the height of plant would be enhaneed with inerease of the radiation dose ranged in this experiment.1he young bulbs were more sensitive to the radiation than mature ones.According to the survival rate.the height of plant and the result of regression analysis,15—20 Gy can be considered as proper radiation dose for youn g bulbs an d 25—30 Gy f0r ma ture ones in the stan dard of LDso and GR5o values.
