大白菜与羽衣甘蓝杂交胚的发育及其离体培养

摘要: 改良Ogura胞质大白菜与羽衣甘蓝的种间杂合胚胎存在明显发育障碍,败育始于球形胚阶段,表现为胚乳细胞化及过度增生;而对照大白菜与甘蓝型油菜杂合胚胎发育至早心形期胚乳仍呈游离核状态。大白菜与羽衣甘蓝杂合胚珠离体培养的有效取材时间为授粉后6~17d,其中9~11d最佳。液体振荡培养可显著促进胚珠离体成苗,花器等营养添加物未见明显效果。

Abstract: There were obvious obstacles of interspecific hybrid embryo growth between CMS Brassica campestris ssp. Pekinensis × B. oleracea var. acephala. Embryos collapse began at sphere stage, with endosperm nuclei being surrounded by walls. Also, endosperms inflated extremely. However, heart embryos between B. campestris ssp. pekinensis×B. napus grew well, surrounding by endosperm nucleus. The available time of ovule rescue culture in vitro was from 6 to 17 days after pollination, and the best time of which was from 9 to 11 days. Surge culture in liquid media improved plantlets notably. Otherwise, flower subsidiaries and other nutritious additions had not shown remarkable influence.
