平欧杂交榛组织培养与快速繁殖技术研究

Studies on tissue culture and rapid propagation of hybrid hazelnut

摘要: 以达维、金铃、玉坠、平顶黄、薄壳红等5个平欧杂交榛品种茎段和根蘖苗茎段为外植体,进行适宜基本培养基的筛选、不定芽的诱导和快速繁殖研究。结果表明:(1)分别以茎段、根蘖苗茎段为外植体进行初代培养,污染率为75%以上和20%以下,外植体存活率为15%以下和65%以上,增殖倍数为低于0.3和高于1.5;(2)在NRM、DKW、WPM、NN、MS等5种基本培养基中,NRM培养基最适宜平欧杂交榛品种试管苗的生长,其茎长和平均芽数高于其它培养基;(3)培养基NRM+5 mg·L-1 6-BA+0.01 mg·L-1 IBA+32.21 mg·L-1 腐胺+29.05 mg·L-1亚精胺+ 10.10 mg·L-1精胺有利于外植体芽体的萌发与生长;(4)试管苗蘸1 g·L-1IBA 20 s进行试管外扦插生根,15 d后生根率可达95%以上,移栽成活率在97%以上。

Abstract: Stem segment and root tiller explants of hybrid hazelnut cultivar Dawei, Jinling, Yuzhui, Pingdinghuang and Bokehong were used to select proper basic medium, establish buds inducement and rapid propagation procedure. The results showed that: (1) during primary culture process, stem segment and root tiller explants were cultured in DKW medium, and the contaminate rate was above 75% and below 20%, survive rate of explants were below 15% and above 65%, propagation times were below 0.3 and above 1.5 respectively. (2) Among basic medium NRM、DKW、WPM、NN and MS, shoot length and auxiliary buds per shoot in NRM medium were higher apparently compared with other medium. (3) Medium NRM+5 mg·L-1 6-BA+0.01 mg·L-1 IBA+64.42 mg·L-1 putrescine+58.10 mg·L-1 spermidine + 20.20 mg·L-1 spermine were profitable to promote buds germination and growth. (4) tube plantlet could root in 15 d and rooting rate reached over 95% when its end dipped in 1 g·L-1 IBA for 20 seconds, and its roots were strong. Transplant survive rate was over 97%.
