西瓜抗病毒基因zym-CH的比较图谱定位

Comparative Mapping of Virus Resistant Gene zym-CH in Watermelon

摘要: 以西瓜抗ZYMV-CH(小西葫芦黄花叶病毒中国株系,Zucchini Yellow Mosaic Virus-CHINA)野生种质P.I.595203和感病自交系98R为试材构建F2群体和F2:3群体,采用BSA法筛选与抗病基因zym-CH连锁的分子标记,并利用以F2S8群体(P.I.296341-FR×97103)构建的西瓜参考遗传图谱进行zym-CH的比较定位。试验结果表明:在参考遗传图谱已经定位的RAPD和SSR标记中,RAPD标记b13-1400与zym-CH的遗传距离为25.6 cM;SSR标记MCPI-14和 MCPI-26zym-CH的遗传距离分别为57 cM和87 cM,比较分析发现这些标记在两个图谱上的分布呈共线性关系。根据zym-CH与标记b13-1400、MCPI-14和MCPI-26之间的遗传关系,将西瓜抗病毒病基因zym-CH初步定位在西瓜参考遗传图谱3号连锁群105 cM处。

Abstract: The watermelon germplasm P.I.595203 resistant to ZYMV-CH ( Zucchini Yellow Mosaic Virus-CHINA ) and 98R susceptible to ZYMV-CH were used as the parents to construct F2 population and F2:3 population. The markers linked to the resistant gene zym-CH were filtered by BSA strategy.The reference genetic map constructed basing on the F2S8 population(P.I.296341-FR×97103)was used for comparative mapping of zym-CH gene. The results showed that b13-1400, MCPI-14 and MCPI-26 were linked to zym-CH gene with the genetic distance of 25.6 cM, 57cM and 87cM respectively. The resistant gene zym-CH was located on 105 cM of the linkage group 3 by the collinearity relationship of zym-CH, b13-1400, MCPI-14 and MCPI-26 in the two linkage maps.
